The roles of NOSs in the pathogenesis of heart failure have been described in pharmacological studies with NOS inhibitors. Recently, genetically engineered animals have been used. We have generated mice in which all 3 NOS isoforms are completely disrupted (triple n/i/eNOS(-/-) mice). Morphological, echocardiographic, and hemodynamic analysis were performed in wild-type, singly nNOS(-/-), iNOS(-/-), eNOS(-/-), and triple n/i/eNOS(-/-) mice. Importantly, significant left ventricular (LV) hypertrophy and diastolic dysfunction was noted only in n/i/eNOS(-/-) mice, and those pathology was similar to diastolic heart failure in humans. Finally, treatment with an angiotensin II type 1 (AT1) receptor blocker, significantly prevented those abnormalities. These results provide the evidence that AT1 receptor pathway plays a center role in the pathogenesis of cardiac disorders in the n/i/eNOS(-/-) mice. Our studies with triple n/i/eNOS(-/-) mice provide pivotal insights into an understanding of the pathophysiology of NOSs in human heart failure.","is_dataset_classified":null,"base_score":2.5649493574615367,"endowment":2.5649493574615367,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"23774658","pmcid":null,"openalex_id":"https://openalex.org/W2066360730","authors":[],"funders":[],"total_grants":0,"fwci":1.3294,"citation_percentile":0.79959816,"influential_citations":0,"citation_trend":[{"year":2014,"count":5},{"year":2015,"count":2},{"year":2016,"count":1},{"year":2017,"count":1},{"year":2018,"count":1},{"year":2019,"count":2}],"oa_status":"bronze","license":null,"oa_locations":[{"url":"https://www.jstage.jst.go.jp/article/juoeh/35/2/35_147/_pdf","host_type":"journal"},{"url":"https://www.jstage.jst.go.jp/article/juoeh/35/2/35_147/_pdf","host_type":"publisher"},{"url":"https://doi.org/10.7888/juoeh.35.147","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/23774658","host_type":"repository"}],"fields_of_study":["Nitric Oxide and Endothelin Effects","Renin-Angiotensin System Studies","Receptor Mechanisms and Signaling","Animals","Disease Models, Animal","Heart Failure","Heart Failure, Diastolic","Mice","Mice, Knockout","Nitric Oxide Synthase","Nitric Oxide Synthase Type I","Nitric Oxide Synthase Type II","Nitric Oxide Synthase Type III"],"mesh_terms":["Animals","Disease Models, Animal","Heart Failure","Mice, Knockout","Nitric Oxide Synthase","Mice","Nitric Oxide Synthase Type II","Nitric Oxide Synthase Type I","Nitric Oxide Synthase Type III","Heart Failure, Diastolic"],"keywords":["Enos","Heart failure","Endothelial NOS","Pathogenesis","Nitric oxide synthase","Nitric oxide","Internal medicine","Angiotensin II receptor type 1","NOS1","Medicine","Endocrinology","Nitric Oxide Synthase Type III","Gene isoform","Angiotensin II","Receptor","Biology","Gene","Biochemistry"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Good health and well-being"}],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-03T03:44:28.393757Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
