For colon carcinogenesis, however, constitutive signaling through the canonical Wnt pathway is not a singular event. Here we review how canonical Wnt signaling is modulated by intracellular LEF/TCF composition and location, the action of different Wnt ligands, and the secretion of Wnt inhibitory molecules. We also review the contributions of non-canonical Wnt signaling and other distinct pathways in the tumor micro environment that cross-talk to the canonical Wnt pathway and thereby influence colon cancer progression. These 'non-APC' aspects of Wnt signaling are considered in relation to the development of potential agents for the treatment of patients with colon cancer. Regulatory pathways that influence Wnt signaling highlight how it might be possible to design therapies that target a network of signals beyond that of APC and β-catenin.","is_dataset_classified":null,"base_score":5.225746673713202,"endowment":5.225746673713202,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"21483657","pmcid":"PMC3072659","openalex_id":"https://openalex.org/W2006485696","authors":[],"funders":[{"funder_name":"NCI NIH HHS","grant_id":"K24 CA082450","title":null},{"funder_name":"NCI NIH HHS","grant_id":"R01 CA096878","title":null},{"funder_name":"NCI NIH HHS","grant_id":"R01 CA108697","title":null},{"funder_name":"National Institutes of Health","grant_id":"5K24CA082450-03","title":"Patient oriented Research--Colon CA &Cancer Immunology"}],"total_grants":4,"fwci":5.476,"citation_percentile":0.96926038,"influential_citations":0,"citation_trend":[{"year":2012,"count":14},{"year":2013,"count":11},{"year":2014,"count":17},{"year":2015,"count":16},{"year":2016,"count":22},{"year":2017,"count":16},{"year":2018,"count":20},{"year":2019,"count":20},{"year":2020,"count":16},{"year":2021,"count":8},{"year":2022,"count":8},{"year":2023,"count":5},{"year":2024,"count":2},{"year":2025,"count":6},{"year":2026,"count":1}],"oa_status":"gold","license":"CC BY NC SA","oa_locations":[{"url":"http://carcinogenesis.com/images/pdf/JCarcinog_2011_10_1_5_78111.pdf","host_type":"journal"},{"url":"http://carcinogenesis.com/images/pdf/JCarcinog_2011_10_1_5_78111.pdf","host_type":"publisher"},{"url":"https://doi.org/10.4103/1477-3163.78111","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/21483657","host_type":"repository"},{"url":"https://doaj.org/article/248dfac391e348cfa6a8a539b4c6c655","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/3072659","host_type":"repository"},{"url":"http://www.biomedcentral.com/1477-3163/10/5","host_type":"BioMedCentral"},{"url":"http://www.biomedcentral.com/1477-3163/10/5/abstract","host_type":"BioMedCentral"},{"url":"http://www.biomedcentral.com/content/pdf/1477-3163-10-5.pdf","host_type":"BioMedCentral"},{"url":"https://europepmc.org/articles/PMC3072659","host_type":"Europe_PMC"},{"url":"https://europepmc.org/articles/PMC3072659?pdf=render","host_type":"Europe_PMC"},{"url":"http://dx.doi.org/10.4103/1477-3163.78111","host_type":""},{"url":"https://dx.doi.org/10.4103/1477-3163.78111","host_type":""}],"fields_of_study":["Wnt/β-catenin signaling in development and cancer","Genetic factors in colorectal cancer","Digestive system and related health","0301 basic medicine","0303 health sciences","03 medical and health sciences"],"mesh_terms":[],"keywords":["Wnt signaling pathway","Medicine","Carcinogenesis","Cancer research","Colorectal cancer","Signal transduction","Biology","Genetics","Internal medicine","Cancer","Wnt signaling","Adenomatous Polyposis Coli Gene","Colon Carcinogenesis","Review Article"],"sdg_mappings":[{"sdg_number":3,"sdg_label":"3.
