Recent publications showed that tenascin-C expression by cancer lesions predicts tumor growth, metastasis, and angiogenesis, suggesting tenascin-C as a potential therapeutic target. Currently there is no noninvasive method to determine tumoral tenascin-C expression in vivo. To address the need for an agent to image and quantify tenascin-C, we report the development of a radioactive PET tracer based on a tenascin-C-specific single-stranded DNA aptamer (tenascin-C aptamer). METHODS: Tenascin-C aptamer was radiolabeled with (18)F and (64)Cu. PET imaging studies for the evaluation of tumor uptake and pharmacokinetics of tenascin-C aptamer were performed in comparison to a nonspecific scrambled aptamer (Sc aptamer). RESULTS: The labeled tenascin-C aptamer provided clear visualization of tenascin-C-positive but not tenascin-C-negative tumors. The uptake of tenascin-C aptamer was significantly higher than that of Sc aptamer in tenascin-C-positive tumors. The labeled tenascin-C aptamer had fast clearance from the blood and other nonspecific organs through the kidneys, resulting in high tumor contrast. CONCLUSION: Our data suggest that suitably labeled tenascin-C aptamer can be used as a PET tracer to image tumor expression of tenascin-C with a high tumor-to-background ratio and might provide insightful and personalized medical data that will help determine appropriate treatment and monitoring.","is_dataset_classified":null,"base_score":4.442651256490317,"endowment":4.442651256490317,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"25698784","pmcid":"PMC5226408","openalex_id":"https://openalex.org/W2073540937","authors":[],"funders":[{"funder_name":"Intramural NIH HHS","grant_id":"Z99 EB999999","title":null},{"funder_name":"Intramural NIH HHS","grant_id":"ZIA EB000073","title":null}],"total_grants":2,"fwci":5.8436,"citation_percentile":0.96990465,"influential_citations":0,"citation_trend":[{"year":2015,"count":7},{"year":2016,"count":6},{"year":2017,"count":6},{"year":2018,"count":12},{"year":2019,"count":5},{"year":2020,"count":7},{"year":2021,"count":12},{"year":2022,"count":6},{"year":2023,"count":4},{"year":2024,"count":5},{"year":2025,"count":10},{"year":2026,"count":4}],"oa_status":"bronze","license":null,"oa_locations":[{"url":"http://jnm.snmjournals.org/content/56/4/616.full.pdf","host_type":"journal"},{"url":"http://jnm.snmjournals.org/content/56/4/616.full.pdf","host_type":"publisher"},{"url":"https://syndication.highwire.org/content/doi/10.2967/jnumed.114.149484","host_type":"publisher"},{"url":"https://doi.org/10.2967/jnumed.114.149484","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/25698784","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/5226408","host_type":"repository"}],"fields_of_study":["Cell Adhesion Molecules Research","Cellular Mechanics and Interactions","Proteoglycans and glycosaminoglycans research","Animals","Aptamers, Nucleotide","Cell Line, Tumor","Copper Radioisotopes","DNA, Single-Stranded","Diagnostic Imaging","Female","Fluorine Radioisotopes","Humans","Mice","Mice, Nude","Microscopy, Fluorescence","Neoplasm Transplantation","Nucleic Acid Conformation","Nucleotides","Positron-Emission Tomography","Radioactive Tracers","Radiopharmaceuticals","Tenascin"],"mesh_terms":["Animals","Copper Radioisotopes","Diagnostic Imaging","DNA, Single-Stranded","Female","Fluorine Radioisotopes","Humans","Mice, Nude","Microscopy, Fluorescence","Neoplasm Transplantation","Nucleic Acid Conformation","Nucleotides","Radioactive Tracers","Tenascin","Radiopharmaceuticals","Cell Line, Tumor","Positron-Emission Tomography","Mice","Aptamers, Nucleotide"],"keywords":["Tenascin C","Aptamer","Tenascin","Chemistry","Cancer research","Extracellular matrix","Molecular biology","Pathology","Medicine","Biology","Biochemistry","PET","Tenascin-c","Dna Aptamer"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Good health and well-being"}],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-03T00:47:56.989612Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
