CryoEM with exhaustive focused classification allowed detection of endogenous proteins associated with porcine kidney V-ATPase. An extra C subunit was found in ∼3% of complexes, whereas ∼1.6% of complexes bound mEAK-7, a protein with proposed roles in dauer formation in nematodes and mTOR signaling in mammals. High-resolution cryoEM of porcine kidney V-ATPase with recombinant mEAK-7 showed that mEAK-7’s TLDc domain interacts with V-ATPase’s stator, whereas its C-terminal α helix binds V-ATPase’s rotor. This crosslink would be expected to inhibit rotary catalysis. However, unlike the yeast TLDc protein Oxr1p, exogenous mEAK-7 does not inhibit V-ATPase and mEAK-7 overexpression in cells does not alter lysosomal or phagosomal pH. Instead, cryoEM suggests that the mEAK-7:V-ATPase interaction is disrupted by ATP-induced rotation of the rotor. Comparison of Oxr1p and mEAK-7 binding explains this difference. These results show that V-ATPase binding by TLDc domain proteins can lead to effects ranging from strong inhibition to formation of labile interactions that are sensitive to the enzyme’s activity.
","journal":"Life Science Alliance","year":2022,"id":679618,"datarank":0.5244761342199721,"base_score":3.4965075614664802,"endowment":3.4965075614664802,"self_citation_contribution":0.5244761342199721,"citation_network_contribution":0.0,"self_endowment_contribution":0.5244761342199721,"citer_contribution":0.0,"corpus_percentile":null,"corpus_rank":null,"citation_count":32,"citer_count":0,"citers_with_citation_signal":0,"citers_with_endowment":0,"datacite_reuse_total":0,"is_dataset":false,"is_dataset_confidence":null,"is_data_producer":false,"deposit_databanks":null,"is_oa":false,"file_count":0,"downloads":0,"has_version_chain":false,"published_date":null,"fair_score":null,"fair_percentile":null,"algorithm_id":"datarank_citation_only_1hop_v6","ranking_scope":"data_only","authors":[{"id":1775701,"name":"Yazan M Abbas","orcid":null,"position":1,"is_corresponding":false},{"id":1241598,"name":"Jing Wu","orcid":"0000-0001-6592-320X","position":2,"is_corresponding":false},{"id":495234,"name":"Di Wu","orcid":"0000-0002-3238-6388","position":3,"is_corresponding":false},{"id":1775702,"name":"Kristine A Keon","orcid":"0000-0001-8185-3803","position":4,"is_corresponding":false},{"id":1775703,"name":"Geoffrey G Hesketh","orcid":"0000-0002-5570-7615","position":5,"is_corresponding":false},{"id":1775704,"name":"Stephanie A Bueler","orcid":null,"position":6,"is_corresponding":false},{"id":44860,"name":"Anne‐Claude Gingras","orcid":"0000-0002-6090-4437","position":7,"is_corresponding":false},{"id":168648,"name":"Carol V Robinson","orcid":null,"position":8,"is_corresponding":false},{"id":325535,"name":"Sergio Grinstein","orcid":"0000-0002-0795-4160","position":9,"is_corresponding":false},{"id":98514,"name":"John L.
