{"doi":"10.2460/ajvr.2002.63.728","title":"Evaluation of in vitro chemosensitivity of vaccine-associated feline sarcoma cell lines to vincristine and paclitaxel","abstract":"<jats:title>Abstract</jats:title>\n\t\t\t\t<jats:p>\n\t\t\t\t\t<jats:bold>Objective</jats:bold>—To determine the in vitro sensitivity of 4\nvaccine-associated feline sarcoma (VAFS) cell lines to\nthe chemotherapeutic agents vincristine and paclitaxel.</jats:p>\n\t\t\t\t<jats:p>\n\t\t\t\t\t<jats:bold>Sample Population</jats:bold>—Cell lines derived from 4 VAFS\nspecimens.</jats:p>\n\t\t\t\t<jats:p>\n\t\t\t\t\t<jats:bold>Procedure</jats:bold>—Cell lines were cultured in vitro and individually\nexposed to various concentrations of vincristine\nand paclitaxel. Survival was estimated after\n24 and 72 hours of exposure to each drug, and the\ndrug concentrations that resulted in 50 and 90%\nreduction in number of viable cells (IC<jats:sub>50</jats:sub> and IC<jats:sub>90</jats:sub>,\nrespectively) were calculated.</jats:p>\n\t\t\t\t<jats:p>\n\t\t\t\t\t<jats:bold>Results</jats:bold>—Both vincristine and paclitaxel had significant\ndose-dependent effects on the viability of the\nVAFS cell lines. After 72 hours of drug exposure, the\nIC<jats:sub>50</jats:sub> and IC<jats:sub>90</jats:sub> of vincristine for the 4 cell lines were\nbetween 0.005 to 0.039 µg/ml and 0.045 to 1.027\nµg/ml, respectively. The IC<jats:sub>50</jats:sub> and IC<jats:sub>90</jats:sub> values for paclitaxel\nwere between 0.037 to 0.092 µg/ml and 2.450\nto 15.413 µg/ml, respectively.</jats:p>\n\t\t\t\t<jats:p>\n\t\t\t\t\t<jats:bold>Conclusions</jats:bold>—Results of pharmacokinetic studies on\nvincristine and paclitaxel in other species suggest that\nconcentrations greater than the IC<jats:sub>50</jats:sub> values may be\npossible for both drugs in feline patients as well. The\ndrug concentrations at which viable cell numbers\nwere reduced by 90% may also be attained in vivo for\nsome cases, but detailed information is needed\nregarding the distribution, concentration, duration of\navailability, and toxicity of various drugs in cats.\nCarefully chosen combinations of antineoplastic\nagents need to be screened to identify treatment protocols\nthat may be further evaluated clinically for the\ntreatment of VAFS.
