In the current study, we searched the literature in different databases for eligible studies. The risk ratio (RR) was measured where p<0.05 was statistically significant. The ACS patients with either one or two CYP2C19 LoF alleles who underwent PCI, treated with clopidogrel were correlated with a significantly escalated risk of MACE compared with noncarriers (RR: 1.53, 95% CI: 1.39–1.69, p < 0.00001), driven by CV death (RR: 1.88, 95% CI: 1.18–3.01, p = 0.008), MI (RR: 1.67, 95% CI: 1.21–2.31, p = 0.002) and ST (RR: 1.90, 95% CI: 1.27–2.84, p = 0.002). Patients with two CYP2C19 LoF alleles were correlated with significantly greater risk of MACE compared with noncarriers (RR: 3.91, 95% CI: 2.78–5.50, p < 0.00001). Further analysis revealed that the risk of MACE was markedly significant in Asian patients (RR: 2.02, 95% CI: 1.67–2.44, p < 0.00001) and was comparatively low significance in western patients (RR: 1.35, 95% CI: 1.20–1.52, p < 0.00001). There was no significantly different bleeding events in patients with CYP2C19 LoF alleles compared with noncarriers (RR: 0.99, 95% CI: 0.85–1.15, p = 0.87). The ACS patients inheriting CYP2C19 LoF alleles, who underwent PCI and were treated with clopidogrel were correlated with significantly increased risk of MACE compared with noncarriers.","is_dataset_classified":null,"base_score":3.367295829986474,"endowment":3.367295829986474,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"35042400","pmcid":null,"openalex_id":"https://openalex.org/W4205092754","authors":[],"funders":[],"total_grants":0,"fwci":2.4436,"citation_percentile":0.89528024,"influential_citations":0,"citation_trend":[{"year":2022,"count":4},{"year":2023,"count":10},{"year":2024,"count":8},{"year":2025,"count":6}],"oa_status":"closed","license":null,"oa_locations":[{"url":"https://www.tandfonline.com/doi/pdf/10.2217/pgs-2021-0098","host_type":"publisher"},{"url":"https://doi.org/10.2217/pgs-2021-0098","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/35042400","host_type":"repository"}],"fields_of_study":["Antiplatelet Therapy and Cardiovascular Diseases","Diabetes Treatment and Management","Lipoproteins and Cardiovascular Health"],"mesh_terms":["Loss of Function Mutation","Clopidogrel","Alleles","Humans","Myocardial Infarction","Platelet Aggregation Inhibitors","Acute Coronary Syndrome","Percutaneous Coronary Intervention","Cytochrome P-450 CYP2C19"],"keywords":["Clopidogrel","Medicine","Acute coronary syndrome","Percutaneous coronary intervention","CYP2C19","Internal medicine","Cardiology","Meta-analysis","Adverse effect","Aspirin","Myocardial infarction","Major Adverse Cardiovascular Events","Cyp2c19 Loss-of-function Alleles"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Good health and well-being"}],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-09T16:29:53.616678Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
