One of the approaches to block the virus spreading is inhibiting viral sialidase (neuraminidase). This enzyme cleaves the sialic acid link between the newly formed virions and the host cell surface liberating the virions from the cell and maintaining the cycle of infection. Viral neuraminidases appear therefore as attractive therapeutic targets for preventing further spread of influenza infection. Compared to ion channel blockers that were the first approved anti-influenza drugs, neuraminidase inhibitors are well tolerated and target both influenza A and B viruses. Moreover, neuraminidase/sialidase inhibitors may be useful for managing some other human pathologies, such as cancer. In this review, we discuss the available knowledge on neuraminidase or sialidase inhibitors, their design, clinical application, and the current challenges.","is_dataset_classified":null,"base_score":4.143134726391533,"endowment":4.143134726391533,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"30349196","pmcid":"PMC6186905","openalex_id":"https://openalex.org/W2896331548","authors":[],"funders":[],"total_grants":0,"fwci":3.7561,"citation_percentile":0.94231116,"influential_citations":0,"citation_trend":[{"year":2018,"count":1},{"year":2019,"count":4},{"year":2020,"count":11},{"year":2021,"count":9},{"year":2022,"count":7},{"year":2023,"count":5},{"year":2024,"count":12},{"year":2025,"count":12},{"year":2026,"count":1}],"oa_status":"gold","license":"cc-by-nc","oa_locations":[{"url":"https://www.dovepress.com/getfile.php?fileID=45181","host_type":"journal"},{"url":"https://www.dovepress.com/getfile.php?fileID=45181","host_type":"publisher"},{"url":"https://doi.org/10.2147/dddt.s176220","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/30349196","host_type":"repository"},{"url":"https://www.dovepress.com/inhibition-of-sialidase-activity-as-a-therapeutic-approach-peer-reviewed-article-DDDT","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/6186905","host_type":"repository"},{"url":"https://europepmc.org/articles/PMC6186905","host_type":"Europe_PMC"},{"url":"https://europepmc.org/articles/PMC6186905?pdf=render","host_type":"Europe_PMC"}],"fields_of_study":["Influenza Virus Research Studies","Monoclonal and Polyclonal Antibodies Research","Glycosylation and Glycoproteins Research","Animals","Antiviral Agents","Enzyme Inhibitors","Humans","Influenza A virus","Influenza B virus","Influenza, Human","Neuraminidase"],"mesh_terms":["Animals","Antiviral Agents","Enzyme Inhibitors","Humans","Influenza, Human","Neuraminidase","Influenza A virus","Influenza B virus"],"keywords":["Neuraminidase","Sialidase","Sialic acid","Virology","Pandemic","Virus","Enzyme","Zanamivir","Biology","Microbiology","Chemistry","Medicine","Coronavirus disease 2019 (COVID-19)","Biochemistry","Disease","Infectious disease (medical specialty)","Drug design","Influenza","neuraminidase inhibitor"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Good health and well-being"}],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-07-29T15:11:51.545505Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
