With the completion of both the human and Drosophila genomes, the revelation that we are more similar to this simple organism than previously suspected was realised. 75% of human genetic disease genes have clear homologues in the fly. By utilising an array of genetic tools available to disrupt or misexpress these proteins, it is now feasible to perform large-scale genetic screens in Drosophila to identify other members of a particular human genetic pathway. This review outlines some of the reasons Drosophila is a useful tool for the discovery of therapeutic targets, covers some of the tools available to manipulate this organism and discusses specific examples of how to use Drosophila as a genetic test tube for revealing proteins which act in a common pathway.","is_dataset_classified":null,"base_score":3.784189633918261,"endowment":3.784189633918261,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"12223075","pmcid":null,"openalex_id":"https://openalex.org/W2139780906","authors":[],"funders":[],"total_grants":0,"fwci":0.303,"citation_percentile":0.4664259,"influential_citations":0,"citation_trend":[{"year":2012,"count":3},{"year":2013,"count":4},{"year":2014,"count":2},{"year":2015,"count":2},{"year":2016,"count":1},{"year":2017,"count":3},{"year":2018,"count":3},{"year":2020,"count":2},{"year":2021,"count":1},{"year":2023,"count":1},{"year":2024,"count":2},{"year":2025,"count":1},{"year":2026,"count":2}],"oa_status":"closed","license":null,"oa_locations":[{"url":"http://www.tandfonline.com/doi/pdf/10.1517/14728222.6.3.387","host_type":"publisher"},{"url":"https://doi.org/10.1517/14728222.6.3.387","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/12223075","host_type":"repository"}],"fields_of_study":["Neurobiology and Insect Physiology Research","Insect Resistance and Genetics","CRISPR and Genetic Engineering","Animals","Caenorhabditis elegans","Disease Models, Animal","Drosophila Proteins","Drosophila melanogaster","Drug Design","Gene Expression Regulation","Genes, Insect","Genetic Diseases, Inborn","Genetic Techniques","Humans","Mice","Models, Animal","Nervous System Diseases","Phenotype","Rats","Saccharomyces cerevisiae","Species Specificity"],"mesh_terms":["Animals","Disease Models, Animal","Drosophila melanogaster","Gene Expression Regulation","Genetic Techniques","Humans","Nervous System Diseases","Phenotype","Saccharomyces cerevisiae","Species Specificity","Drug Design","Caenorhabditis elegans","Genes, Insect","Models, Animal","Drosophila Proteins","Genetic Diseases, Inborn","Mice","Rats"],"keywords":["Drosophila melanogaster","Model organism","Organism","Biology","Drosophila (subgenus)","Genetic screen","Gene","Computational biology","Genetics","Genome","Function (biology)","Human disease","Drug discovery","Phenotype","Bioinformatics"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Good health and well-being"}],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-07-30T02:13:36.086080Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
