The association of NOD2 mutations with a number of inflammatory pathologies, including Crohn disease (CD), Graft-versus-host disease (GVHD), and Blau syndrome, highlights its pivotal role in host-pathogen interactions and inflammatory response. Stimulation of NOD2 by its ligand (muramyl dipeptide) activates pro-inflammatory pathways such as nuclear factor-κB (NF-κB), mitogen-activated protein kinases (MAPKs), and Caspase-1. A loss of NOD2 function may result in a failure in the control of microbial infection, thereby initiating systemic responses and aberrant inflammation. Because the ligand of Nod2 is conserved in both gram-positive and gram-negative bacteria, NOD2 detects a wide variety of microorganisms. Furthermore, current literature evidences that NOD2 is also able to control viruses' and parasites' infections. In this review, we present and discuss recent developments about the role of NOD2 in shaping the gut commensal microbiota and pathogens, including bacteria, viruses, and parasites, and the mechanisms by which Nod2 mutations participate in disease occurrence.","is_dataset_classified":null,"base_score":5.117993812416755,"endowment":5.117993812416755,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"28253332","pmcid":"PMC5333895","openalex_id":"https://openalex.org/W2591843498","authors":[],"funders":[{"funder_name":"French National Research Agency (ANR)","grant_id":"ANR-11-IDEX-0005","title":null}],"total_grants":1,"fwci":10.8486,"citation_percentile":0.99184402,"influential_citations":0,"citation_trend":[{"year":2017,"count":8},{"year":2018,"count":25},{"year":2019,"count":18},{"year":2020,"count":19},{"year":2021,"count":15},{"year":2022,"count":19},{"year":2023,"count":27},{"year":2024,"count":9},{"year":2025,"count":17},{"year":2026,"count":9}],"oa_status":"gold","license":"cc-by","oa_locations":[{"url":"https://journals.plos.org/plospathogens/article/file?id=10.1371/journal.ppat.1006177&type=printable","host_type":"journal"},{"url":"https://journals.plos.org/plospathogens/article/file?id=10.1371/journal.ppat.1006177&type=printable","host_type":"publisher"},{"url":"https://doi.org/10.1371/journal.ppat.1006177","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/28253332","host_type":"repository"},{"url":"https://hal.science/hal-01602910","host_type":"repository"},{"url":"https://doaj.org/article/26c70da68711401c9a3a6f8b2601eda1","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/5333895","host_type":"repository"},{"url":"https://europepmc.org/articles/PMC5333895","host_type":"Europe_PMC"},{"url":"https://europepmc.org/articles/PMC5333895?pdf=render","host_type":"Europe_PMC"},{"url":"http://dx.doi.org/10.1371/journal.ppat.1006177","host_type":""},{"url":"https://dx.doi.org/10.1371/journal.ppat.1006177","host_type":""},{"url":"https://hal.science/hal-01602910v1","host_type":""},{"url":"https://hal.science/hal-01602910v1/document","host_type":""},{"url":"http://prodinra.inra.fr/record/405725","host_type":""},{"url":"https://doi.org/https://doi.org/10.1371/journal.ppat.1006177","host_type":""}],"fields_of_study":["Viral gastroenteritis research and epidemiology","Mycobacterium research and diagnosis","Bacteriophages and microbial interactions","0301 basic medicine","03 medical and health sciences"],"mesh_terms":["Gastrointestinal Microbiome","Animals","Humans","Intestines","Nod2 Signaling Adaptor Protein","Host-Pathogen Interactions"],"keywords":["NOD2","Muramyl dipeptide","Peptidoglycan","Biology","Pattern recognition receptor","Immune system","Microbiology","Immunology","Bacteria","Genetics","Immunity","Innate immune system","570","QH301-705.5","flore commensale","Nod2 Signaling Adaptor Protein","610","microbiote digestif","Review","Animals","Humans","Biology (General)","commensal flora","interaction hôte pathogène","[SDV.MHEP.HEG]Life Sciences [q-bio]/Human health and pathology/Hépatology and Gastroenterology","RC581-607","[SDV.MHEP.HEG] Life Sciences [q-bio]/Human health and pathology/Hépatology and Gastroenterology","Gastrointestinal Microbiome","Hépatologie et Gastroentérologie","Intestines","Host-Pathogen Interactions","Hépatology and Gastroenterology","réaction inflammatoire","Immunologic diseases.
