Entry by fusion with the endosomal membrane requires the fusion loop (FL, residues 507-560) of the EBOV surface glycoprotein to undergo a pH-dependent conformational change. To find the pH trigger for this reaction we mutated multiple conserved histidines and charged and uncharged hydrophilic residues in the FL and measured their activity by liposome fusion and cell entry of virus-like particles. The FL location in the membrane was assessed by NMR using soluble and lipid-bound paramagnetic relaxation agents. While we could not identify a single residue to be alone responsible for pH triggering, we propose that a distributed pH effect over multiple residues induces the conformational change that enhances membrane insertion and triggers the fusion activity of the EBOV FL.","is_dataset_classified":null,"base_score":2.9444389791664403,"endowment":2.9444389791664403,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"27023721","pmcid":"PMC4811418","openalex_id":"https://openalex.org/W2314363083","authors":[],"funders":[{"funder_name":"NIAID NIH HHS","grant_id":"R01 AI030557","title":null},{"funder_name":"NIGMS NIH HHS","grant_id":"T32 GM008136","title":null},{"funder_name":"NIGMS NIH HHS","grant_id":"T32 GM080186","title":null}],"total_grants":3,"fwci":1.2867,"citation_percentile":0.78677864,"influential_citations":0,"citation_trend":[{"year":2016,"count":1},{"year":2017,"count":4},{"year":2019,"count":1},{"year":2020,"count":2},{"year":2021,"count":2},{"year":2023,"count":4},{"year":2024,"count":2},{"year":2025,"count":1},{"year":2026,"count":1}],"oa_status":"gold","license":"cc-by","oa_locations":[{"url":"https://journals.plos.org/plosone/article/file?id=10.1371/journal.pone.0152527&type=printable","host_type":"journal"},{"url":"https://journals.plos.org/plosone/article/file?id=10.1371/journal.pone.0152527&type=printable","host_type":"publisher"},{"url":"http://dx.plos.org/10.1371/journal.pone.0152527","host_type":"publisher"},{"url":"https://doi.org/10.1371/journal.pone.0152527","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/27023721","host_type":"repository"},{"url":"https://doaj.org/article/9da00d73077e4807855666d69143221e","host_type":"repository"},{"url":"https://figshare.com/articles/dataset/The_Roles_of_Histidines_and_Charged_Residues_as_Potential_Triggers_of_a_Conformational_Change_in_the_Fusion_Loop_of_Ebola_Virus_Glycoprotein/3152542","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/4811418","host_type":"repository"},{"url":"https://europepmc.org/articles/PMC4811418","host_type":"Europe_PMC"},{"url":"https://europepmc.org/articles/PMC4811418?pdf=render","host_type":"Europe_PMC"}],"fields_of_study":["Viral Infections and Outbreaks Research","Viral Infections and Vectors","Viral gastroenteritis research and epidemiology","Amino Acid Sequence","Circular Dichroism","Ebolavirus","Histidine","Hydrogen-Ion Concentration","Magnetic Resonance Spectroscopy","Molecular Sequence Data","Protein Structure, Secondary","Recombinant Fusion Proteins","Sequence Alignment","Structure-Activity Relationship","Viral Envelope Proteins"],"mesh_terms":["Amino Acid Sequence","Circular Dichroism","Histidine","Hydrogen-Ion Concentration","Molecular Sequence Data","Magnetic Resonance Spectroscopy","Recombinant Fusion Proteins","Structure-Activity Relationship","Viral Envelope Proteins","Sequence Alignment","Protein Structure, Secondary","Ebolavirus"],"keywords":["Endosome","Lipid bilayer fusion","Conformational change","Ebola virus","Biophysics","Chemistry","Glycoprotein","Lipid bilayer","Residue (chemistry)","Fusion","Viral membrane","Liposome","Membrane","Viral entry","Biochemistry","Virus","Biology","Cell","Virology","Viral replication","Viral envelope"],"sdg_mappings":[],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[{"name":"pdb"}],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-02T19:33:21.185589Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
