Furthermore, a variety of protein partners have been identified that can interact with and modulate DAT function. In this study we show that DJ-1 can potentially modulate DAT function. Co-expression of DAT and DJ-1 in HEK-293T cells leads to an increase in [3H] dopamine uptake that does not appear to be mediated by increased total DAT expression but rather through an increase in DAT cell surface localization. In addition, through a series of GST affinity purifications and co-immunoprecipitations, we provide evidence that the DAT can be found in a complex with DJ-1, which involve distinct regions within both DAT and DJ-1. Using in vitro binding experiments we also show that this complex can be formed in part by a direct interaction between DAT and DJ-1. Co-expression of a mini-gene that can disrupt the DAT/DJ-1 complex appears to block the increase in [3H] dopamine uptake by DJ-1. Mutations in DJ-1 have been linked to familial forms of Parkinson's disease, yet the normal physiological function of DJ-1 remains unclear. Our study suggests that DJ-1 may also play a role in regulating dopamine levels by modifying DAT activity.","is_dataset_classified":null,"base_score":2.995732273553991,"endowment":2.995732273553991,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"26305376","pmcid":"PMC4549284","openalex_id":"https://openalex.org/W1891167896","authors":[],"funders":[{"funder_name":"Natural Sciences and Engineering Research Council of Canada","grant_id":"unidentified","title":"unidentified"}],"total_grants":1,"fwci":1.3219,"citation_percentile":0.80126656,"influential_citations":0,"citation_trend":[{"year":2017,"count":3},{"year":2018,"count":3},{"year":2019,"count":1},{"year":2020,"count":4},{"year":2021,"count":1},{"year":2022,"count":1},{"year":2023,"count":1},{"year":2025,"count":3},{"year":2026,"count":2}],"oa_status":"gold","license":"cc-by","oa_locations":[{"url":"https://journals.plos.org/plosone/article/file?id=10.1371/journal.pone.0136641&type=printable","host_type":"journal"},{"url":"https://journals.plos.org/plosone/article/file?id=10.1371/journal.pone.0136641&type=printable","host_type":"publisher"},{"url":"http://dx.plos.org/10.1371/journal.pone.0136641","host_type":"publisher"},{"url":"https://doi.org/10.1371/journal.pone.0136641","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/26305376","host_type":"repository"},{"url":"https://figshare.com/articles/dataset/_A_Physical_Interaction_between_the_Dopamine_Transporter_and_DJ_1_Facilitates_Increased_Dopamine_Reuptake_/1520134","host_type":"repository"},{"url":"https://doaj.org/article/7c424bfee7cd438eb8aef9b8cc6adbee","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/4549284","host_type":"repository"},{"url":"http://summit.sfu.ca/item/16010","host_type":"repository"},{"url":"https://europepmc.org/articles/PMC4549284","host_type":"Europe_PMC"},{"url":"https://europepmc.org/articles/PMC4549284?pdf=render","host_type":"Europe_PMC"},{"url":"http://dx.doi.org/10.1371/journal.pone.0136641","host_type":""},{"url":"https://dx.doi.org/10.1371/journal.pone.0136641","host_type":""},{"url":"https://summit.sfu.ca/item/16010","host_type":""}],"fields_of_study":["Parkinson's Disease Mechanisms and Treatments","Nuclear Receptors and Signaling","Nerve injury and regeneration","0301 basic medicine","03 medical and health sciences","0303 health sciences"],"mesh_terms":["Protein Deglycase DJ-1","Cell Membrane","Dopamine","Gene Expression Regulation","Humans","Mutation","Parkinson Disease","Protein Binding","Oncogene Proteins","Immunoprecipitation","Intracellular Signaling Peptides and Proteins","Dopamine Plasma Membrane Transport Proteins","HEK293 Cells","Dopaminergic Neurons"],"keywords":["Dopamine transporter","Dopamine","Reuptake","Dopaminergic","Dopamine Plasma Membrane Transport Proteins","HEK 293 cells","Transporter","Cell biology","Extracellular","Chemistry","Biology","Neuroscience","Biochemistry","Gene","Receptor","Serotonin","Co-immunoprecipitation","Science","Protein Deglycase DJ-1","Transfection","Humans","Immunoprecipitation","Small interfering RNAs","Neurons","Oncogene Proteins","Dopaminergic Neurons","Q","Cell Membrane","R","Intracellular Signaling Peptides and Proteins","Parkinson Disease","Complementary DNA","HEK293 Cells","Neuronal differentiation","Gene Expression Regulation","Mutation","Medicine","Dopaminergics","Research Article","Protein Binding"],"sdg_mappings":[{"sdg_number":2,"sdg_label":"2.
