Due to their central role in biological function, protein interactions also control the mechanisms leading to healthy and diseased states in organisms. Diseases are often caused by mutations affecting the binding interface or leading to biochemically dysfunctional allosteric changes in proteins. Therefore, protein interaction networks can elucidate the molecular basis of disease, which in turn can inform methods for prevention, diagnosis, and treatment. In this chapter, we will describe the computational approaches to predict and map networks of protein interactions and briefly review the experimental methods to detect protein interactions. We will describe the application of protein interaction networks as a translational approach to the study of human disease and evaluate the challenges faced by these approaches.","is_dataset_classified":null,"base_score":5.641907070938114,"endowment":5.641907070938114,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"23300410","pmcid":"PMC3531279","openalex_id":"https://openalex.org/W1998222399","authors":[],"funders":[{"funder_name":"NCI NIH HHS","grant_id":"K22 CA143148","title":null},{"funder_name":"NCI NIH HHS","grant_id":"1K22CA143148","title":null},{"funder_name":"NLM NIH HHS","grant_id":"R01 LM009722","title":null},{"funder_name":"National Institutes of Health","grant_id":"5K22CA143148-03","title":"Protein domains as molecular units to unravel functional pathways of cancer genes"},{"funder_name":"National Institutes of Health","grant_id":"5R01LM009722-09","title":"Informatic Profiling of Clinically Relevant Mutation"},{"funder_name":"Intramural NIH HHS","grant_id":"","title":null}],"total_grants":6,"fwci":3.8046,"citation_percentile":0.94373264,"influential_citations":0,"citation_trend":[{"year":2013,"count":7},{"year":2014,"count":8},{"year":2015,"count":16},{"year":2016,"count":11},{"year":2017,"count":14},{"year":2018,"count":26},{"year":2019,"count":27},{"year":2020,"count":19},{"year":2021,"count":25},{"year":2022,"count":28},{"year":2023,"count":35},{"year":2024,"count":26},{"year":2025,"count":29},{"year":2026,"count":9}],"oa_status":"gold","license":"cc-by","oa_locations":[{"url":"https://journals.plos.org/ploscompbiol/article/file?id=10.1371/journal.pcbi.1002819&type=printable","host_type":"journal"},{"url":"https://journals.plos.org/ploscompbiol/article/file?id=10.1371/journal.pcbi.1002819&type=printable","host_type":"publisher"},{"url":"http://dx.plos.org/10.1371/journal.pcbi.1002819","host_type":"publisher"},{"url":"https://doi.org/10.1371/journal.pcbi.1002819","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/23300410","host_type":"repository"},{"url":"https://doaj.org/article/3bf8ba09e7f9462c9033a2666a68735c","host_type":"repository"},{"url":"https://figshare.com/articles/dataset/Chapter_4_Protein_Interactions_and_Disease_/115127","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/3531279","host_type":"repository"},{"url":"https://europepmc.org/articles/PMC3531279","host_type":"Europe_PMC"},{"url":"https://europepmc.org/articles/PMC3531279?pdf=render","host_type":"Europe_PMC"},{"url":"http://dx.doi.org/10.1371/journal.pcbi.1002819","host_type":""},{"url":"https://dx.doi.org/10.1371/journal.pcbi.1002819","host_type":""}],"fields_of_study":["Bioinformatics and Genomic Networks","Computational Drug Discovery Methods","Microbial Metabolic Engineering and Bioproduction","0301 basic medicine","0303 health sciences","03 medical and health sciences"],"mesh_terms":["Disease","Humans","Mutation","Protein Binding"],"keywords":["Computational biology","Protein–protein interaction","Allosteric regulation","Function (biology)","Protein Interaction Networks","Biology","Protein function","Disease","Bioinformatics","Genetics","Medicine","Gene","QH301-705.5","Mutation","Humans","Biology (General)","Education","Protein Binding"],"sdg_mappings":[{"sdg_number":3,"sdg_label":"3.
