We previously reported that the intranasal delivery of a Th1-inducing antigen promoted Th2-dominated responses, rather than the expected Th1 responses. Thus, we proposed that when pulmonary T cell priming is induced, the lung microenvironment might intrinsically favor the generation of Th2 types of responses. To establish a potential mechanism for such preferential priming, we examined the initial interactions between antigens and resident antigen-presenting cells (APCs) within the lung. We show that intranasally delivered antigens are preferentially taken up and can be presented to antigen-specific T cells by a resident population of CD11cbright APCs. Most of these antigen-loaded APCs remained within lung tissues, and migration into secondary lymphoid organs was not crucial for T cell priming to occur within the pulmonary tract. Furthermore, these pulmonary APCs demonstrated a marked expression of IL-6 and IL-10 within hours of antigen uptake, suggesting that resident tissue APCs have the capacity to promote Th2 T cell differentiation in situ.","is_dataset_classified":null,"base_score":5.062595033026967,"endowment":5.062595033026967,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"23304386","pmcid":null,"openalex_id":"https://openalex.org/W4243641777","authors":[],"funders":[],"total_grants":0,"fwci":4.9617,"citation_percentile":0.96242411,"influential_citations":0,"citation_trend":[{"year":2012,"count":5},{"year":2013,"count":7},{"year":2014,"count":5},{"year":2015,"count":4},{"year":2016,"count":4},{"year":2017,"count":3},{"year":2020,"count":4},{"year":2021,"count":2},{"year":2022,"count":1},{"year":2023,"count":3},{"year":2024,"count":4},{"year":2025,"count":1},{"year":2026,"count":2}],"oa_status":"closed","license":null,"oa_locations":[{"url":"http://www.jci.org/articles/view/16109/files/pdf","host_type":"publisher"},{"url":"https://doi.org/10.1172/jci0216109","host_type":"journal"}],"fields_of_study":["Immune Cell Function and Interaction","Asthma and respiratory diseases","Immunotherapy and Immune Responses"],"mesh_terms":[],"keywords":["Priming (agriculture)","Antigen","Immunology","Antigen-presenting cell","Population","T cell","Biology","Cell biology","Immune system","Medicine"],"sdg_mappings":[],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-07-29T17:27:15.583052Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
