Such proteins must be properly folded and posttranslationally modified before exit from the organelle. Proper protein folding and modification requires molecular chaperone proteins as well as an ER environment conducive for these reactions. When ER lumenal conditions are altered or chaperone capacity is overwhelmed, the cell activates signaling cascades that attempt to deal with the altered conditions and restore a favorable folding environment. Such alterations are referred to as ER stress, and the response activated is the unfolded protein response (UPR). When the UPR is perturbed or not sufficient to deal with the stress conditions, apoptotic cell death is initiated. This review will examine UPR signaling that results in cell protective responses, as well as the mechanisms leading to apoptosis induction, which can lead to pathological states due to chronic ER stress.</jats:p>","journal":"Physiology","year":2007,"id":22956,"datarank":12.200326418443552,"base_score":6.163314804034641,"endowment":6.163314804034641,"self_citation_contribution":0.9244972206051963,"citation_network_contribution":11.275829197838355,"self_endowment_contribution":0.9244972206051963,"citer_contribution":11.275829197838355,"corpus_percentile":null,"corpus_rank":null,"citation_count":474,"citer_count":200,"citers_with_citation_signal":200,"citers_with_endowment":200,"datacite_reuse_total":2,"is_dataset":false,"is_dataset_confidence":null,"is_oa":false,"file_count":0,"downloads":0,"has_version_chain":false,"published_date":null,"algorithm_id":"datarank_citation_only_1hop_v6","ranking_scope":"data_only","authors":[{"id":142615,"name":"Tracy Teodoro","orcid":null,"position":1,"is_corresponding":false},{"id":142616,"name":"Allen Volchuk","orcid":null,"position":2,"is_corresponding":false},{"id":142614,"name":"Elida Lai","orcid":null,"position":0,"is_corresponding":false}],"reference_count":0,"raw_metadata":{"has_enrichment":true,"base_score":6.163314804034641,"endowment":6.163314804034641,"datacite_reuse_total":2,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"17557940","pmcid":null,"openalex_id":"https://openalex.org/W2161604602","authors":[],"funders":[],"total_grants":0,"fwci":8.7422,"citation_percentile":0.98633533,"influential_citations":40,"citation_trend":[{"year":2012,"count":54},{"year":2013,"count":47},{"year":2014,"count":42},{"year":2015,"count":27},{"year":2016,"count":36},{"year":2017,"count":25},{"year":2018,"count":24},{"year":2019,"count":18},{"year":2020,"count":24},{"year":2021,"count":24},{"year":2022,"count":21},{"year":2023,"count":16},{"year":2024,"count":10},{"year":2025,"count":8}],"oa_status":"closed","license":null,"oa_locations":[{"url":"https://www.physiology.org/doi/pdf/10.1152/physiol.00050.2006","host_type":"publisher"},{"url":"https://doi.org/10.1152/physiol.00050.2006","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/17557940","host_type":"repository"}],"fields_of_study":["Endoplasmic Reticulum Stress and Disease","Autophagy in Disease and Therapy","Pancreatic function and diabetes","Biology","Medicine","Animals","Apoptosis","Endoplasmic Reticulum, Rough","Humans","Protein Folding","Protein Processing, Post-Translational","Signal Transduction"],"mesh_terms":["Animals","Humans","Protein Processing, Post-Translational","Signal Transduction","Apoptosis","Protein Folding","Endoplasmic Reticulum, Rough"],"keywords":["Endoplasmic reticulum","Unfolded protein response","Cell biology","Chaperone (clinical)","Protein folding","Organelle","Signal transduction","Chemical chaperone","Cell","Secretory protein","Secretory pathway","Biology","Chemistry","Secretion","Biochemistry","Golgi apparatus","Medicine"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Life in Land"}],"linked_datasets":[{"doi":"10.6084/m9.figshare.13213033","title":"Additional file 1 of Acupuncture protects against cerebral ischemia–reperfusion injury via suppressing endoplasmic reticulum stress-mediated autophagy and apoptosis","publisher":"figshare","resource_type":"Image"},{"doi":"10.6084/m9.figshare.13213033.v1","title":"Additional file 1 of Acupuncture protects against cerebral ischemia–reperfusion injury via suppressing endoplasmic reticulum stress-mediated autophagy and apoptosis","publisher":"figshare","resource_type":"Image"}],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-06-07T17:16:18.642617Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
