The main purpose of this investigation was to explore the effect of miR-15b on lipid metabolism in mammary epithelial cells, along with the underlying mechanisms. miR-15b was overexpressed or inhibited by miRNA mimics or inhibitors; subsequently, lipid formation in mammary epithelial cells, and proteins related to lipid metabolism, were investigated. Through overexpression or inhibition of miR-15b expression, the current investigation found that miR-15b downregulates lipid metabolism in mammary epithelial cells and is expressed differentially at various stages of mouse and goat mammary gland development. Inhibition of miR-15b expression increased lipid content in mammary epithelial cells through elevation of the lipid synthesis enzyme fatty acid synthetase (FASN), and overexpression of miR-15b reduced lipid content in mammary epithelial cells with decreasing levels of FASN. Moreover, the steroid hormones estradiol and progesterone decreased miR-15b expression with a subsequent increase in lipid formation in mammary epithelial cells. The expression of miR-15b was lower during lactation and negatively correlated with lipid synthesis proteins, which suggests that it may be involved in lipid synthesis and milk production. miR-15b might be a useful target for altering lipid production and milk yield.
","journal":"American Journal of Physiology-Cell Physiology","year":2018,"id":640614,"datarank":0.5416376868966337,"base_score":3.6109179126442243,"endowment":3.6109179126442243,"self_citation_contribution":0.5416376868966337,"citation_network_contribution":0.0,"self_endowment_contribution":0.5416376868966337,"citer_contribution":0.0,"corpus_percentile":null,"corpus_rank":null,"citation_count":36,"citer_count":0,"citers_with_citation_signal":0,"citers_with_endowment":0,"datacite_reuse_total":0,"is_dataset":false,"is_dataset_confidence":null,"is_data_producer":false,"deposit_databanks":null,"is_oa":false,"file_count":0,"downloads":0,"has_version_chain":false,"published_date":null,"fair_score":null,"fair_percentile":null,"algorithm_id":"datarank_citation_only_1hop_v6","ranking_scope":"data_only","authors":[{"id":765590,"name":"Yong Zhao","orcid":"0000-0003-3423-2718","position":1,"is_corresponding":false},{"id":695273,"name":"Shuai Yu","orcid":"0000-0001-5563-2492","position":2,"is_corresponding":false},{"id":1664993,"name":"Yanan Hao","orcid":null,"position":3,"is_corresponding":false},{"id":303851,"name":"Pengfei Zhang","orcid":"0000-0002-9458-5067","position":4,"is_corresponding":false},{"id":1664994,"name":"Yanni Feng","orcid":null,"position":5,"is_corresponding":false},{"id":765585,"name":"Hongfu Zhang","orcid":"0000-0003-0177-8773","position":6,"is_corresponding":false},{"id":765586,"name":"Dongxue Ma","orcid":"0000-0003-1140-3676","position":7,"is_corresponding":false},{"id":756065,"name":"Jing Liu","orcid":"0000-0002-3588-2969","position":8,"is_corresponding":false},{"id":496262,"name":"Ming Cheng","orcid":"0000-0002-1581-6382","position":9,"is_corresponding":false},{"id":897155,"name":"Lan Li","orcid":"0000-0003-1255-5102","position":10,"is_corresponding":false},{"id":246136,"name":"Wei Shen","orcid":"0000-0002-8099-8258","position":11,"is_corresponding":false},{"id":1664995,"name":"Hongfang Cao","orcid":null,"position":12,"is_corresponding":false},{"id":211885,"name":"Qiang Li","orcid":"0009-0008-9097-8585","position":13,"is_corresponding":false},{"id":766244,"name":"Lingjiang Min","orcid":null,"position":14,"is_corresponding":false},{"id":1664987,"name":"Meiqiang Chu","orcid":null,"position":0,"is_corresponding":false}],"reference_count":0,"raw_metadata":{"has_enrichment":true,"resolved":true,"title":"miR-15b negatively correlates with lipid metabolism in mammary epithelial cells","abstract":"
Mammary epithelial cells are regulated by steroid hormones, growth factors, and even microRNAs. miR-15b has been found to regulate lipid metabolism in adipocytes; however, its effects on lipid metabolism in mammary epithelial cells, the cells of lipid synthesis and secretion, are as yet unknown.
