In the absence of substrate, Lon adopts a left-handed, \"open\" spiral organization with autoinhibited proteolytic active sites. Upon the addition of substrate, Lon undergoes a reorganization to assemble an enzymatically active, right-handed \"closed\" conformer with active protease sites.
