{"doi":"10.1111/cga.12011","title":"Wnt11 expression in rat dental pulp and promotional effects of <scp>W</scp>nt signaling on odontoblast differentiation","abstract":"<jats:title>Abstract</jats:title><jats:p>Congenital anomalies of wingless‐type mouse mammary tumor virus (<jats:styled-content style=\"fixed-case\">MMTV)</jats:styled-content> integration site family (<jats:styled-content style=\"fixed-case\">W</jats:styled-content>nt) are frequently accompanied with tooth and dentin abnormality. The aim of this study was to investigate the effects of <jats:styled-content style=\"fixed-case\">W</jats:styled-content>nt signaling on odontoblast differentiation of mouse dental papilla cells (<jats:styled-content style=\"fixed-case\">MDPs</jats:styled-content>). Mouse dental papilla cells were cultured in α‐modified minimum essential medium containing 10% fetal bovine serum and antibiotics. Odontoblast differentiation was induced by bone morphogenic protein 2 (<jats:styled-content style=\"fixed-case\">BMP2</jats:styled-content>), and the expression of odontoblast‐specific markers and Wnt‐related signaling molecules was analyzed by real‐time reverse transcription‐polymerase chain reaction and immunohistochemistry. Odontoblast differentiation was evaluated by <jats:italic>dentin sialophosphoprotein</jats:italic> (<jats:italic><jats:styled-content style=\"fixed-case\">D</jats:styled-content>spp</jats:italic>) and dentin matrix protein (<jats:styled-content style=\"fixed-case\">DMP</jats:styled-content>) 1 expression. Localization of β‐catenin in <jats:styled-content style=\"fixed-case\">MDPs</jats:styled-content> was detected by immunocytochemistry using an anti‐β‐catenin antibody. <jats:italic><jats:styled-content style=\"fixed-case\">D</jats:styled-content>spp</jats:italic> expression in <jats:styled-content style=\"fixed-case\">MDPs</jats:styled-content> was upregulated in the presence of <jats:styled-content style=\"fixed-case\">BMP2</jats:styled-content>. <jats:italic><jats:styled-content style=\"fixed-case\">W</jats:styled-content>nt5a</jats:italic>, <jats:italic><jats:styled-content style=\"fixed-case\">W</jats:styled-content>nt11</jats:italic>, <jats:italic><jats:styled-content style=\"fixed-case\">L</jats:styled-content>ef1</jats:italic> and <jats:italic><jats:styled-content style=\"fixed-case\">T</jats:styled-content>cf4</jats:italic> expression was upregulated in <jats:styled-content style=\"fixed-case\">BMP2</jats:styled-content>‐treated <jats:styled-content style=\"fixed-case\">MDPs</jats:styled-content>. <jats:styled-content style=\"fixed-case\">W</jats:styled-content>nt11 expression was detected in rat dental pulp <jats:italic>in vivo</jats:italic>, and particularly strong expression of <jats:styled-content style=\"fixed-case\">W</jats:styled-content>nt11 was detected in odontoblasts. Enhanced <jats:italic><jats:styled-content style=\"fixed-case\">D</jats:styled-content>spp</jats:italic> and <jats:styled-content style=\"fixed-case\">DMP1</jats:styled-content> expression and alkaline phosphatase activity induced by <jats:styled-content style=\"fixed-case\">BMP2</jats:styled-content> were completely negated by the <jats:styled-content style=\"fixed-case\">W</jats:styled-content>nt antagonist: <jats:styled-content style=\"fixed-case\">IWR</jats:styled-content>‐1‐endo treatment. Nuclear translocation of β‐catenin observed in <jats:styled-content style=\"fixed-case\">BMP2</jats:styled-content>‐treated <jats:styled-content style=\"fixed-case\">MDPs</jats:styled-content> was also negated by <jats:styled-content style=\"fixed-case\">IWR</jats:styled-content>‐1‐endo treatment.
