{"doi":"10.1111/acer.14617","title":"Speaker Abstracts","abstract":"Our laboratory has been investigating the effects of developmental ethanol exposure in fetal alcohol spectrum disorder (FASD) models on astrocyte functions affecting neuronal development with an emphasis on astrocyte-released extracellular matrix (ECM). The Aldh1 l1-EGFP-Rpl10a mouse model expresses a modified ribosomal protein Rpl10a with an eGFP tag in cells expressing Aldh1 l1 (a highly specific astrocytic marker) that allow to isolate astrocyte specific RNA using the translating ribosome affinity purification (TRAP) method. As ribosomes are assembled in the nucleoli, the Ald-h1 l1-EGFP-Rpl10a mouse line has GFP fluorescence within the nucleus (where nucleoli are located) that enable the isolation of astrocyte-specific nuclei by Fluorescent-Activated Cell Sorting (FACS). The sorted astrocyte nuclei from brain tissue allows the analysis of astrocyte-specific gene transcription (mRNA present in the nuclei) and of DNA methylation. Using the TRAP method followed by qPCR we investigated hippocampal astrocyte translation in vivo at postnatal day (PD) 7 after alcohol vapor or air (control) exposure between PD2 and PD7. We found laminins ?2, ?5, ?2, and c1 (Lama2; Lama5, Lamb2, Lamc1) to be enriched in hippocampal astrocytes compared to the whole hippocampus. Adamts9, Mmp14, Mmp15, Plat, Plau, Timp1, Timp3, and Timp4, genes encoding for extracellular matrix proteases, are enriched in hippocampal astrocytes compared to the whole hippocampus. We also found that, Lama2, Lama5, Adamts4, Adamts5, Adamts9, and Dnmt3b (encoding for DNA methyltransferase 3b) are upregulated selectively in astrocytes by neonatal alcohol exposure. We have begun analysis on astrocyte nuclei collected by FACS from mouse cortex showing enrichment of astrocyte specific RNA as well as enrichment of RNAs known to reside in the nucleus. In addition, we have characterized astrocyte DNA collected by FACS which showed increased DNA methylation near transcriptional start sites (TSSs) of non-astrocytic markers, as expected for silenced genes, and decreased DNA methylation near TSSs of astrocyte markers, as expected for actively transcribed genes.
