{"doi":"10.1101/2021.03.15.435527","title":"Inflammatory signals are sufficient to elicit TOX expression in mouse and human CD8 T cells","abstract":"Abstract T cell receptor (TCR) stimulation leads to expression of the transcription factor TOX. Prolonged TCR signaling, such as encountered during chronic infections or in tumors, leads to sustained TOX expression, which induces a state of exhaustion or dysfunction. While CD8 memory T cells (T mem ) in specific pathogen-free laboratory mice typically do not express TOX, functional human T mem show heterogeneous TOX expression levels. Whether TCR-independent mechanisms can alter TOX expression in human and murine T mem has not been defined. We report that human and mouse T mem increase TOX expression following stimulation with inflammatory cytokines IL-12, IL-15, and IL-18. TOX and PD-1 expression patterns often appear to be directly correlated, however, we found that TOX is not necessary for cytokine-driven expression of PD-1. Together, these observations highlight that inflammation is sufficient to alter TOX and PD-1 expression and that the signals regulating TOX expression appear well conserved in human and murine T mem .","journal":"bioRxiv (Cold Spring Harbor Laboratory)","year":2021,"id":220837,"datarank":0.0,"base_score":0.0,"endowment":0.0,"self_citation_contribution":0.0,"citation_network_contribution":0.0,"self_endowment_contribution":0.0,"citer_contribution":0.0,"corpus_percentile":null,"corpus_rank":null,"citation_count":1,"citer_count":0,"citers_with_citation_signal":0,"citers_with_endowment":0,"datacite_reuse_total":0,"is_dataset":false,"is_dataset_confidence":0.951,"is_data_producer":false,"deposit_databanks":null,"is_oa":true,"file_count":0,"downloads":0,"has_version_chain":false,"published_date":"2021-01-01","fair_score":null,"fair_percentile":null,"algorithm_id":"datarank_citation_only_1hop_v6","ranking_scope":"data_only","authors":[{"id":669446,"name":"Jacqueline Berner","orcid":"0000-0002-9056-8745","position":1,"is_corresponding":false},{"id":655546,"name":"Alexis K Taber","orcid":"0000-0002-7012-3001","position":2,"is_corresponding":false},{"id":271014,"name":"Dietmar Zehn","orcid":"0000-0003-1393-8527","position":3,"is_corresponding":false},{"id":272541,"name":"Martin Prlic","orcid":"0000-0002-0685-9321","position":4,"is_corresponding":false},{"id":430770,"name":"Nicholas J.
