Despite aggressive treatment modalities, the median survival duration for patients with glioblastoma multiforme (GBM), the highest grade malignant glioma, has not improved significantly over past decades. One promising approach to deal with GBM is the inactivation of proteins essential for survival or progression of glioma cells by means of RNA interference (RNAi) techniques. A likely candidate for an RNAi therapy of gliomas is the inhibitor of apoptosis protein survivin. Survivin is involved in 2 main cellular processes-cell division and inhibition of apoptosis. We show here that stable RNAi of survivin induced polyploidy, apoptosis, and impaired proliferation of human U343-MG, U373-MG, H4, and U87-MG cells and of primary glioblastoma cells. Proteome profiler arrays using U373-MG cells identified a novel set of differentially expressed genes upon RNAi-mediated survivin knockdown. In particular, the death receptor TRAIL R2/DR5 was strongly upregulated in survivin-depleted glioma cells, inducing an enhanced cytotoxic response of allogeneic human NK cells. Moreover, an experimental in vivo therapy using polyethylenimine (PEI)/siRNA complexes for survivin knockdown efficiently blocked tumor growth of established subcutaneous U373-MG tumors and enhanced survival of NMRI(nu/nu) mice orthopically transplanted with U87-MG cells. We conclude that survivin is functionally relevant in gliomas and that PEI-mediated exogenous delivery of siRNA targeting survivin is a promising strategy for glioblastoma therapy.","is_dataset_classified":null,"base_score":4.248495242049359,"endowment":4.248495242049359,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"21788344","pmcid":"PMC3177660","openalex_id":"https://openalex.org/W2159271922","authors":[],"funders":[],"total_grants":0,"fwci":2.9177,"citation_percentile":0.91893703,"influential_citations":0,"citation_trend":[{"year":2012,"count":6},{"year":2013,"count":10},{"year":2014,"count":7},{"year":2015,"count":5},{"year":2016,"count":4},{"year":2017,"count":8},{"year":2018,"count":4},{"year":2019,"count":5},{"year":2020,"count":2},{"year":2021,"count":5},{"year":2022,"count":6},{"year":2024,"count":2},{"year":2025,"count":3},{"year":2026,"count":1}],"oa_status":"hybrid","license":"cc-by-nc","oa_locations":[{"url":"https://academic.oup.com/neuro-oncology/article-pdf/13/10/1074/3590931/nor098.pdf","host_type":"journal"},{"url":"https://academic.oup.com/neuro-oncology/article-pdf/13/10/1074/3590931/nor098.pdf","host_type":"publisher"},{"url":"http://academic.oup.com/neuro-oncology/article-pdf/13/10/1074/3590931/nor098.pdf","host_type":"publisher"},{"url":"https://doi.org/10.1093/neuonc/nor098","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/21788344","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/3177660","host_type":"repository"},{"url":"https://europepmc.org/articles/PMC3177660","host_type":"Europe_PMC"},{"url":"https://europepmc.org/articles/PMC3177660?pdf=render","host_type":"Europe_PMC"}],"fields_of_study":["RNA Interference and Gene Delivery","Cell death mechanisms and regulation","Advanced biosensing and bioanalysis techniques","Animals","Apoptosis","Blotting, Western","Brain Neoplasms","Cell Separation","Flow Cytometry","Fluorescent Antibody Technique","Genetic Therapy","Glioma","Humans","In Situ Nick-End Labeling","Inhibitor of Apoptosis Proteins","Mice","Protein Array Analysis","RNA Interference","RNA, Small Interfering","Real-Time Polymerase Chain Reaction","Survivin","Transduction, Genetic","Xenograft Model Antitumor Assays"],"mesh_terms":["Survivin","Animals","Brain Neoplasms","Cell Separation","Flow Cytometry","Fluorescent Antibody Technique","Glioma","Humans","Transduction, Genetic","Blotting, Western","Genetic Therapy","Apoptosis","In Situ Nick-End Labeling","Xenograft Model Antitumor Assays","RNA Interference","RNA, Small Interfering","Protein Array Analysis","Inhibitor of Apoptosis Proteins","Mice","Real-Time Polymerase Chain Reaction"],"keywords":["Survivin","Gene knockdown","Glioma","RNA interference","Cancer research","Apoptosis","Small hairpin RNA","Small interfering RNA","U87","Biology","In vivo","Chemistry","Cell culture","Transfection","RNA","Gene"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Industry, innovation and infrastructure"}],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[{"name":"doi"}],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-11T23:49:51.351759Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
