Forty percent of the human genome is comprised of transposable element sequences, and the most abundant cut-and-paste transposons are from the hAT superfamily. We found that the hAT transposase TcBuster from Tribolium castaneum formed filamentous structures, or rodlets, in human tissue culture cells, after gene transfer to adult mice, and ex vivo in cell-free conditions, indicating that host co-factors or cellular structures were not required for rodlet formation. Time-lapsed imaging of GFP-laced rodlets in human cells revealed that they formed quickly in a dynamic process involving fusion and fission. We delayed the availability of the transposon DNA and found that transposition declined after transposase concentrations became high enough for visible transposase rodlets to appear. In combination with earlier findings for maize Ac elements, these results give insight into transposase overproduction inhibition by demonstrating that the appearance of transposase protein structures and the end of active transposition are simultaneous, an effect with implications for genetic engineering and horizontal gene transfer.","is_dataset_classified":null,"base_score":4.787491742782046,"endowment":4.787491742782046,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"27899587","pmcid":"PMC5224482","openalex_id":"https://openalex.org/W2558229157","authors":[],"funders":[{"funder_name":"NIGMS NIH HHS","grant_id":"R25 GM069234","title":null},{"funder_name":"NIDDK NIH HHS","grant_id":"R01 DK093660","title":null},{"funder_name":"BLRD VA","grant_id":"I01 BX002190","title":null},{"funder_name":"BLRD VA","grant_id":"IK2 BX002797","title":null},{"funder_name":"National Institutes of Health","grant_id":"5T35DK007383-41","title":"Vanderbilt Student Research Training Program"},{"funder_name":"National Institutes of Health","grant_id":"2R25GM069234-14","title":"Post-baccalaureate Research Education Program (PREP) (R25)"},{"funder_name":"National Institutes of Health","grant_id":"2T32DK060445-11","title":"Hematology Training Program"},{"funder_name":"National Institutes of Health","grant_id":"5R01DK093660-10","title":"Novel cell therapy for sustained therapeutic protein delivery in vivo"},{"funder_name":"National Institutes of Health","grant_id":"5T32DK062706-08","title":"Research Training in Renal Diseases"}],"total_grants":9,"fwci":35.2765,"citation_percentile":0.99741241,"influential_citations":0,"citation_trend":[{"year":2017,"count":8},{"year":2018,"count":20},{"year":2019,"count":27},{"year":2020,"count":24},{"year":2021,"count":29},{"year":2022,"count":1},{"year":2023,"count":5},{"year":2024,"count":2},{"year":2025,"count":1},{"year":2026,"count":2}],"oa_status":"gold","license":"public-domain","oa_locations":[{"url":"https://academic.oup.com/nar/article-pdf/45/1/353/9249427/gkw1115.pdf","host_type":"journal"},{"url":"https://academic.oup.com/nar/article-pdf/45/1/353/9249427/gkw1115.pdf","host_type":"publisher"},{"url":"http://academic.oup.com/nar/article-pdf/45/1/353/9249427/gkw1115.pdf","host_type":"publisher"},{"url":"https://doi.org/10.1093/nar/gkw1115","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/27899587","host_type":"repository"},{"url":"http://nar.oxfordjournals.org/cgi/content/short/45/1/353","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/5224482","host_type":"repository"},{"url":"https://europepmc.org/articles/PMC5224482","host_type":"Europe_PMC"},{"url":"https://europepmc.org/articles/PMC5224482?pdf=render","host_type":"Europe_PMC"},{"url":"http://dx.doi.org/10.1093/nar/gkw1115","host_type":""},{"url":"https://dx.doi.org/10.1093/nar/gkw1115","host_type":""}],"fields_of_study":["Chromosomal and Genetic Variations","CRISPR and Genetic Engineering","Advanced biosensing and bioanalysis techniques","0301 basic medicine","0303 health sciences","03 medical and health sciences"],"mesh_terms":["Animals","DNA Transposable Elements","Female","Gene Expression Regulation","HeLa Cells","Humans","Luciferases","Male","Tribolium","Genes, Reporter","Insect Proteins","Transposases","Mice","HEK293 Cells","Time-Lapse Imaging","Optical Imaging","Hela Cells"],"keywords":["Transposase","Biology","Transposable element","Transposition (logic)","Genetics","Insertion sequence","Genome","P element","Gene","Male","Tribolium","Nucleic Acid Enzymes","Optical Imaging","Transposases","Time-Lapse Imaging","Mice","HEK293 Cells","Gene Expression Regulation","Genes, Reporter","DNA Transposable Elements","Animals","Humans","Insect Proteins","Female","Luciferases","HeLa Cells"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Life in Land"}],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-04T04:05:10.069200Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
