But the strength of proteins-their versatility, dynamics and interactions-also complicates and hinders systems engineering. Therefore, the design of more sophisticated, multi-component protein systems appears to lag behind, in particular, when compared to the engineering of gene regulatory networks. Yet, synthetic biologists have started to tinker with the information flow through natural signaling networks or integrated protein switches. A successful strategy common to most of these experiments is their focus on modular interactions between protein domains or domains and peptide motifs. Such modular interaction swapping has rewired signaling in yeast, put mammalian cell morphology under the control of light, or increased the flux through a synthetic metabolic pathway. Based on this experience, we outline an engineering framework for the connection of reusable protein interaction devices into self-sufficient circuits. Such a framework should help to 'refacture' protein complexity into well-defined exchangeable devices for predictive engineering. We review the foundations and initial success stories of protein synthetic biology and discuss the challenges and promises on the way from protein- to protein systems design.","is_dataset_classified":null,"base_score":4.6913478822291435,"endowment":4.6913478822291435,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"20385577","pmcid":"PMC2860127","openalex_id":"https://openalex.org/W2025658351","authors":[],"funders":[{"funder_name":"European Commission","grant_id":"201648","title":"PROteomics SPECification in Time and Space"}],"total_grants":1,"fwci":5.2251,"citation_percentile":0.96495135,"influential_citations":0,"citation_trend":[{"year":2012,"count":11},{"year":2013,"count":14},{"year":2014,"count":10},{"year":2015,"count":7},{"year":2016,"count":8},{"year":2017,"count":5},{"year":2018,"count":6},{"year":2019,"count":3},{"year":2020,"count":6},{"year":2021,"count":6},{"year":2022,"count":5},{"year":2023,"count":2},{"year":2024,"count":4},{"year":2025,"count":2},{"year":2026,"count":2}],"oa_status":"gold","license":"other-oa","oa_locations":[{"url":"https://academic.oup.com/nar/article-pdf/38/8/2663/11217954/gkq139.pdf","host_type":"journal"},{"url":"https://academic.oup.com/nar/article-pdf/38/8/2663/11217954/gkq139.pdf","host_type":"publisher"},{"url":"http://academic.oup.com/nar/article-pdf/38/8/2663/11217954/gkq139.pdf","host_type":"publisher"},{"url":"https://doi.org/10.1093/nar/gkq139","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/20385577","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/2860127","host_type":"repository"},{"url":"https://europepmc.org/articles/PMC2860127","host_type":"Europe_PMC"},{"url":"https://europepmc.org/articles/PMC2860127?pdf=render","host_type":"Europe_PMC"},{"url":"http://dx.doi.org/10.1093/nar/gkq139","host_type":""},{"url":"https://dx.doi.org/10.1093/nar/gkq139","host_type":""}],"fields_of_study":["Gene Regulatory Network Analysis","CRISPR and Genetic Engineering","RNA and protein synthesis mechanisms","0301 basic medicine","0303 health sciences","03 medical and health sciences"],"mesh_terms":["DNA","Models, Molecular","Proteins","Protein Engineering","Signal Transduction","Protein Interaction Mapping","Intracellular Signaling Peptides and Proteins","Protein Interaction Domains and Motifs"],"keywords":["Synthetic biology","Biology","Protein engineering","Modular design","Computational biology","Systems biology","Protein design","Component (thermodynamics)","Protein–protein interaction","Metabolic engineering","Biochemical engineering","Computer science","Protein structure","Cell biology","Engineering","Gene","Genetics","Biochemistry","Models, Molecular","Synthetic Biology and Chemistry","Protein Interaction Mapping","Intracellular Signaling Peptides and Proteins","Proteins","Protein Interaction Domains and Motifs","DNA","Signal Transduction"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Industry, innovation and infrastructure"}],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-07-28T14:08:56.586073Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
