Recently, it was shown that additional elements, such as exonic splicing enhancers (ESEs), are essential for the proper splicing of certain exons, in addition to the splice donor and acceptor site sequences; however, the relationship between these cis-elements is still unclear. In this report, we utilize dystrophin exon 19 to analyse the relationship between the ESE and its upstream acceptor site sequences. Dystrophin exon 19, which maintains adequate splicing donor and acceptor consensus sequences, encodes exonic splicing enhancer (dys-ESE19) sequences. Splice pattern analysis, using a minigene reporter expressed in HeLa cells, showed that either a strong polypyrimidine tract (PPT) or a fully active dys-ESE19 is sufficient for proper splicing. Each of these two cis-elements has enough activity for proper exon 19 splicing suggesting that the PPT, which is believed to be an essential cis-element for splicing, is dispensable when the downstream exon contains a strong ESE. This compensation was only seen in living cells but not in 'in vitro splicing'. This suggests the possibility that the previous splicing experiments using an in vitro splicing system could underestimate the activity of ESEs.","is_dataset_classified":null,"base_score":2.639057329615259,"endowment":2.639057329615259,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"18039686","pmcid":null,"openalex_id":"https://openalex.org/W2012651446","authors":[],"funders":[],"total_grants":0,"fwci":0.3997,"citation_percentile":0.6144251,"influential_citations":1,"citation_trend":[{"year":2014,"count":1},{"year":2016,"count":2},{"year":2017,"count":1},{"year":2019,"count":2},{"year":2020,"count":2}],"oa_status":"closed","license":null,"oa_locations":[{"url":"http://academic.oup.com/jb/article-pdf/143/3/303/14113544/mvm227.pdf","host_type":"publisher"},{"url":"https://doi.org/10.1093/jb/mvm227","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/18039686","host_type":"repository"},{"url":"http://jb.oxfordjournals.org/cgi/content/short/mvm227v1","host_type":"repository"},{"url":"http://jb.oxfordjournals.org/cgi/content/short/mvm227v2","host_type":"repository"}],"fields_of_study":["RNA Research and Splicing","Muscle Physiology and Disorders","RNA Interference and Gene Delivery","Biology","Medicine","Alternative Splicing","Base Sequence","Cell Extracts","Cell Nucleus","Dystrophin","Enhancer Elements, Genetic","Exons","Genes, Reporter","HeLa Cells","Humans","Molecular Sequence Data","Mutation","Polyribonucleotides","RNA Splice Sites","Sequence Analysis, DNA","Transfection"],"mesh_terms":["Base Sequence","Cell Extracts","Cell Nucleus","Enhancer Elements, Genetic","Exons","HeLa Cells","Humans","Molecular Sequence Data","Mutation","Polyribonucleotides","Transfection","Dystrophin","Alternative Splicing","Sequence Analysis, DNA","Genes, Reporter","RNA Splice Sites","Hela Cells"],"keywords":["Polypyrimidine tract","RNA splicing","Exon","Exonic splicing enhancer","Polypyrimidine tract-binding protein","Enhancer","Genetics","Computational biology","Biology","Alternative splicing","Gene","RNA","Gene expression"],"sdg_mappings":[],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-07-15T02:34:21.500303Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
