In mammalian cells, mTOR is part of two different kinase complexes: mTORC1 composed of mTOR, raptor and mLST8, and mTORC2 containing mTOR, rictor, sin1 and mLST8. Whereas, mTORC1 is known to be a pivotal regulator of cell size and cell cycle control, the question whether the recently discovered mTORC2 complex is involved in these processes remains elusive. We report here that the mTORC1-mediated consequences on cell cycle and cell size are separable and do not involve effects on mTORC2 activity. However, we show that mTORC2 itself is a potent regulator of mammalian cell size and cell cycle via a mechanism involving the Akt/TSC2/Rheb cascade. Our data are of relevance for the understanding of the molecular development of the many human diseases caused by deregulation of upstream and downstream effectors of mTOR.","is_dataset_classified":null,"base_score":3.9889840465642745,"endowment":3.9889840465642745,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"19505958","pmcid":"PMC2722991","openalex_id":"https://openalex.org/W2154106855","authors":[],"funders":[{"funder_name":"Austrian Science Fund (FWF)","grant_id":"P 18894","title":"The role of tuberin in p27 regulation"}],"total_grants":1,"fwci":4.1854,"citation_percentile":0.94922113,"influential_citations":0,"citation_trend":[{"year":2012,"count":10},{"year":2013,"count":3},{"year":2014,"count":3},{"year":2015,"count":1},{"year":2016,"count":5},{"year":2017,"count":2},{"year":2018,"count":1},{"year":2019,"count":2},{"year":2021,"count":3},{"year":2022,"count":2}],"oa_status":"hybrid","license":"cc-by-nc","oa_locations":[{"url":"https://academic.oup.com/hmg/article-pdf/18/17/3298/8224605/ddp271.pdf","host_type":"journal"},{"url":"https://academic.oup.com/hmg/article-pdf/18/17/3298/8224605/ddp271.pdf","host_type":"publisher"},{"url":"https://academic.oup.com/hmg/article/18/17/3298/2527387/Functional-interaction-of-mammalian-target-of","host_type":"publisher"},{"url":"https://doi.org/10.1093/hmg/ddp271","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/19505958","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/2722991","host_type":"repository"},{"url":"https://europepmc.org/articles/PMC2722991","host_type":"Europe_PMC"},{"url":"https://europepmc.org/articles/PMC2722991?pdf=render","host_type":"Europe_PMC"},{"url":"http://dx.doi.org/10.1093/hmg/ddp271","host_type":""},{"url":"https://dx.doi.org/10.1093/hmg/ddp271","host_type":""}],"fields_of_study":["PI3K/AKT/mTOR signaling in cancer","Mast cells and histamine","Polyamine Metabolism and Applications","0301 basic medicine","0303 health sciences","03 medical and health sciences","Cell Cycle","Cell Line","Cell Size","Humans","Multiprotein Complexes","Protein Kinases","TOR Serine-Threonine Kinases"],"mesh_terms":["Cell Cycle","Cell Line","Humans","Protein Kinases","Multiprotein Complexes","Cell Size","TOR Serine-Threonine Kinases"],"keywords":["mTORC2","mTORC1","RHEB","PI3K/AKT/mTOR pathway","Biology","Cell biology","TSC2","RPTOR","Mechanistic target of rapamycin","Cell cycle","Regulator","Protein kinase B","Cell growth","Effector","Signal transduction","Cell","Genetics","Gene","Multiprotein Complexes","TOR Serine-Threonine Kinases","Humans","Articles","Protein Kinases","Cell Line","Cell Size"],"sdg_mappings":[{"sdg_number":3,"sdg_label":"3.
