Previous studies have shown that histone H4 hyperacetylation is important for acute up-regulation of insulin gene transcription. Until now, only the histone acetyltransferase (HAT) protein p300 has been shown to be involved in this histone H4 acetylation event. In this report we investigated the role of the additional HAT proteins CREB binding protein (CBP), p300/CBP-associated factor (PCAF), and general control of amino-acid synthesis 5 (GCN5) in regulation of glucose-stimulated insulin gene transcription. Utilizing quantitative chromatin immunoprecipitation analysis, we demonstrate that glucose regulates the binding of p300, CBP, PCAF, and GCN5 to the proximal insulin promoter. siRNA-mediated knockdown of each of these HAT proteins revealed that depletion of p300 and CBP leads to a drastic decrease in histone H4 acetylation at the insulin promoter and in insulin gene expression, whereas knockdown of PCAF and GCN5 leads to a more moderate decrease in histone H4 acetylation and insulin gene expression. These data suggest that high glucose mediates the recruitment of p300, CBP, PCAF, and GCN5 to the insulin promoter and that all four HATs are important for insulin gene expression.","is_dataset_classified":null,"base_score":3.091042453358316,"endowment":3.091042453358316,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"21774670","pmcid":"PMC3246418","openalex_id":"https://openalex.org/W2087893369","authors":[],"funders":[{"funder_name":"NIDDK NIH HHS","grant_id":"R01DK067581","title":null},{"funder_name":"NCRR NIH HHS","grant_id":"P20RR020171","title":null}],"total_grants":2,"fwci":0.2819,"citation_percentile":0.63948831,"influential_citations":0,"citation_trend":[{"year":2014,"count":1},{"year":2015,"count":1},{"year":2016,"count":2},{"year":2017,"count":4},{"year":2018,"count":1},{"year":2019,"count":2},{"year":2020,"count":1},{"year":2021,"count":2},{"year":2022,"count":1},{"year":2023,"count":4},{"year":2024,"count":1},{"year":2026,"count":1}],"oa_status":"green","license":"http://www.liebertpub.com/nv/resources-tools/text-and-data-mining-policy/121/","oa_locations":[{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/3246418","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/3246418","host_type":"repository"},{"url":"http://www.liebertpub.com/doi/full-xml/10.1089/dna.2011.1336","host_type":"publisher"},{"url":"http://www.liebertpub.com/doi/pdf/10.1089/dna.2011.1336","host_type":"publisher"},{"url":"https://doi.org/10.1089/dna.2011.1336","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/21774670","host_type":"repository"},{"url":"http://europepmc.org/articles/PMC3246418","host_type":"repository"}],"fields_of_study":["Pancreatic function and diabetes","Metabolism, Diabetes, and Cancer","Genetics and Neurodevelopmental Disorders","Acetylation","Animals","Blotting, Western","CREB-Binding Protein","Cell Line, Tumor","Chromatin Immunoprecipitation","Glucose","Histones","Insulin","Insulinoma","Mice","Promoter Regions, Genetic","Protein Binding","RNA Interference","Reverse Transcriptase Polymerase Chain Reaction","Transcription, Genetic","p300-CBP Transcription Factors","p300-CBP-Associated Factor"],"mesh_terms":["p300-CBP-Associated Factor","Acetylation","Animals","Glucose","Histones","Insulin","Insulinoma","Promoter Regions, Genetic","Protein Binding","Transcription, Genetic","Blotting, Western","Reverse Transcriptase Polymerase Chain Reaction","RNA Interference","Cell Line, Tumor","Chromatin Immunoprecipitation","p300-CBP Transcription Factors","CREB-Binding Protein","Mice"],"keywords":["PCAF","Biology","Histone H4","SAP30","CREB-binding protein","Histone acetyltransferase","P300-CBP Transcription Factors","Chromatin immunoprecipitation","Molecular biology","Promoter","Histone H2A","Histone","Transcription factor","Histone Acetyltransferases","Histone H3","GRB10","Acetylation","Insulin receptor","Gene expression","Insulin","Gene","Genetics","CREB","Insulin resistance","Endocrinology"],"sdg_mappings":[],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-04T14:10:26.467550Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
