MATERIALS AND METHODS: ions) or low-LET ionizing radiation (X-rays) under various conditions to produce varied radical-scavenging effects. pUC18 was then treated sequentially or simultaneously with two bacterial base excision enzymes (glycosylases), namely, endonuclease III and formamidopyrimidine-DNA glycosylase, which convert pyrimidine (or abasic [AP] site) and purine (or AP site) lesions to single-strand breaks (SSB), respectively. Yields of additional SSB or double-strand breaks (DSB) as digestion products were examined after changing the order of enzymatic treatment. RESULTS: There were few differences among the enzymatic treatments, indicating that treatment order did not affect the final yields of additional SSB or DSB formed by glycosylase activity. This suggests that of the total damage, the fraction of clustered damage sites with a persistent base lesion dependent on the order of glycosylase treatment was insignificant if present. CONCLUSION: Base lesion clusters induced by high- or low-LET radiation appear three or more base pairs apart, and are promptly converted to a DSB by glycosylase, regardless of the order of enzymatic treatment.","is_dataset_classified":null,"base_score":2.3978952727983707,"endowment":2.3978952727983707,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"27707033","pmcid":null,"openalex_id":"https://openalex.org/W2528508621","authors":[],"funders":[],"total_grants":0,"fwci":0.8542,"citation_percentile":0.76621913,"influential_citations":0,"citation_trend":[{"year":2017,"count":3},{"year":2018,"count":1},{"year":2019,"count":1},{"year":2020,"count":1},{"year":2021,"count":1},{"year":2022,"count":1},{"year":2024,"count":1},{"year":2026,"count":1}],"oa_status":"green","license":"cc-by-nc-nd","oa_locations":[{"url":"https://repo.qst.go.jp/records/47716","host_type":"repository"},{"url":"https://repo.qst.go.jp/records/47716","host_type":"GREEN"},{"url":"https://repo.qst.go.jp/records/47716","host_type":"repository"},{"url":"https://www.tandfonline.com/doi/pdf/10.1080/09553002.2017.1239849","host_type":"publisher"},{"url":"https://doi.org/10.1080/09553002.2017.1239849","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/27707033","host_type":"repository"},{"url":"https://repo.qst.go.jp/records/48015","host_type":"repository"}],"fields_of_study":["DNA Repair Mechanisms","Effects of Radiation Exposure","Cancer Research and Treatments","Medicine","Biology","Environmental Science","Base Pairing","DNA","DNA Damage","DNA Glycosylases","DNA Repair","Deoxyribonuclease I","Dose-Response Relationship, Radiation","Radiation Dosage"],"mesh_terms":["Deoxyribonuclease I","DNA","DNA Damage","DNA Repair","Dose-Response Relationship, Radiation","Radiation Dosage","Base Pairing","DNA Glycosylases"],"keywords":["DNA glycosylase","AP site","Base excision repair","AP endonuclease","DNA-(apurinic or apyrimidinic site) lyase","DNA repair","Endonuclease","Nucleotide excision repair","MUTYH","DNA","DNA damage","Uracil-DNA glycosylase","Chemistry","Lesion","XRCC1","Biochemistry","Molecular biology","Biology","Medicine","Gene","Pathology","Clustered Dna Damage","High-let Radiation","Base Lesions"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Affordable and clean energy"}],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-07-12T01:00:18.416193Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
