Functional impairment was attributed to over 100-fold loss of CXCR4 binding affinity. Binding to the other CXCL12 receptor, ACKR3, was diminished nearly 500-fold. Deletion of Pro-10 had little effect on CXCL12 binding to the CXCR4 N terminus, a major component of the chemokine-GPCR interface. Replacement of the X residue with the most frequent amino acid at this position (P10Q) had an intermediate effect between WT and P10del in each assay, with ACKR3 having a higher tolerance for this mutation. This work shows that the X residue helps to position the CXCL12 N terminus for optimal docking into the orthosteric pocket of CXCR4 and suggests that the CC/CXC motif contributes directly to receptor selectivity by orienting the chemokine N terminus in a subfamily-specific direction.","journal":"Journal of Biological Chemistry","year":2020,"id":69900,"datarank":0.8076745389709954,"base_score":3.044522437723423,"endowment":3.044522437723423,"self_citation_contribution":0.4566783656585135,"citation_network_contribution":0.3509961733124819,"self_endowment_contribution":0.4566783656585135,"citer_contribution":0.3509961733124819,"corpus_percentile":null,"corpus_rank":null,"citation_count":20,"citer_count":18,"citers_with_citation_signal":17,"citers_with_endowment":17,"datacite_reuse_total":0,"is_dataset":false,"is_dataset_confidence":0.9507,"is_data_producer":true,"deposit_databanks":{"PDB":["2KED","2JYO","4RWS","1EL0","1B50","2FIN","5YAM","6FGP","4ZKC","2MPM","2HCC","2MP1","2L4N","1G91","1EIH","1G2T","2KUM","6CWS","1B2T","1J8I","1MGS","1QNK","2MGS","1ILP","5WDZ","1LV9","1RJT","2N55","5L7M","2HDL"]},"is_oa":true,"file_count":0,"downloads":0,"has_version_chain":false,"published_date":"2020-01-01","fair_score":null,"fair_percentile":null,"algorithm_id":"datarank_citation_only_1hop_v6","ranking_scope":"data_only","authors":[{"id":370581,"name":"Sarah A.
