Still, we know little about the mechanisms that regulate its expression and its activity. In this study, we show that the transcriptional coactivator PGC-1, which is implicated in the control of energy metabolism, regulates ERRalpha at two levels. First, PGC-1 induces the expression of ERRalpha. Consistent with this induction, levels of ERRalpha mRNA in vivo are highest in PGC-1 expressing tissues, such as heart, kidney, and muscle, and up-regulated in response to signals that induce PGC-1, such as exposure to cold. Second, PGC-1 interacts physically with ERRalpha and enables it to activate transcription. Strikingly, we find that PGC-1 converts ERRalpha from a factor with little or no transcriptional activity to a potent regulator of gene expression, suggesting that ERRalpha is not a constitutively active nuclear receptor but rather one that is regulated by protein ligands, such as PGC-1. Our findings suggest that the two proteins act in a common pathway to regulate processes relating to energy metabolism. In support of this hypothesis, adenovirus-mediated delivery of small interfering RNA for ERRalpha, or of PGC-1 mutants that interact selectively with different types of nuclear receptors, shows that PGC-1 can induce the fatty acid oxidation enzyme MCAD (medium-chain acyl-coenzyme A dehydrogenase) in an ERRalpha-dependent manner.","is_dataset_classified":null,"base_score":6.1224928095143865,"endowment":6.1224928095143865,"datacite_reuse_total":2,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"12522104","pmcid":null,"openalex_id":"https://openalex.org/W1990579890","authors":[],"funders":[],"total_grants":0,"fwci":11.5014,"citation_percentile":0.99028425,"influential_citations":0,"citation_trend":[{"year":2012,"count":23},{"year":2013,"count":23},{"year":2014,"count":20},{"year":2015,"count":23},{"year":2016,"count":13},{"year":2017,"count":14},{"year":2018,"count":12},{"year":2019,"count":16},{"year":2020,"count":22},{"year":2021,"count":25},{"year":2022,"count":14},{"year":2023,"count":16},{"year":2024,"count":24},{"year":2025,"count":8},{"year":2026,"count":4}],"oa_status":"hybrid","license":"cc-by","oa_locations":[{"url":"https://doi.org/10.1074/jbc.m212923200","host_type":"journal"},{"url":"https://doi.org/10.1074/jbc.m212923200","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S002192581971268X?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S002192581971268X?httpAccept=text/plain","host_type":"publisher"},{"url":"https://syndication.highwire.org/content/doi/10.1074/jbc.M212923200","host_type":"publisher"},{"url":"https://pubmed.ncbi.nlm.nih.gov/12522104","host_type":"repository"},{"url":"http://www.jbc.org/content/278/11/9013.full.pdf","host_type":"Unpaywall"}],"fields_of_study":["Estrogen and related hormone effects","Adipose Tissue and Metabolism","Nuclear Receptors and Signaling","Acyl-CoA Dehydrogenase","Adenoviridae","Blotting, Western","Cell Line","Cell Nucleus","Fatty Acid Desaturases","Genetic Vectors","Green Fluorescent Proteins","HeLa Cells","Humans","Luminescent Proteins","Mutation","RNA","RNA, Messenger","Receptors, Cytoplasmic and Nuclear","Receptors, Estrogen","Recombinant Fusion Proteins","Reverse Transcriptase Polymerase Chain Reaction","Time Factors","Tissue Distribution","Transcription Factors","Transcription, Genetic","Transfection","Tumor Cells, Cultured","Two-Hybrid System Techniques","Up-Regulation","ERRalpha Estrogen-Related Receptor"],"mesh_terms":["ERRalpha Estrogen-Related Receptor","Adenoviridae","Cell Line","Cell Nucleus","Genetic Vectors","HeLa Cells","Humans","Luminescent Proteins","Mutation","Receptors, Estrogen","Recombinant Fusion Proteins","RNA","RNA, Messenger","Time Factors","Tissue Distribution","Transcription Factors","Transcription, Genetic","Transfection","Tumor Cells, Cultured","Blotting, Western","Up-Regulation","Receptors, Cytoplasmic and Nuclear","Reverse Transcriptase Polymerase Chain Reaction","Two-Hybrid System Techniques","Acyl-CoA Dehydrogenase","Fatty Acid Desaturases","Green Fluorescent Proteins","Hela Cells"],"keywords":["Coactivator","Estrogen-related receptor alpha","Nuclear receptor","Nuclear receptor coactivator 1","Transcription factor","Biology","Receptor","Estrogen receptor","Estrogen-related receptor gamma","Cell biology","Biochemistry","Gene","Genetics"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Affordable and clean energy"}],"linked_datasets":[{"doi":"10.6084/m9.figshare.13364550.v1","title":"Additional file 8 of Estrogen-related receptor alpha directly binds to p53 and cooperatively controls colon cancer growth through the regulation of mitochondrial biogenesis and function","publisher":"figshare","resource_type":"JournalArticle"},{"doi":"10.6084/m9.figshare.13364550","title":"Additional file 8 of Estrogen-related receptor alpha directly binds to p53 and cooperatively controls colon cancer growth through the regulation of mitochondrial biogenesis and function","publisher":"figshare","resource_type":"JournalArticle"}],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-07-23T07:27:29.997533Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
