We previously found that acidic pH increases the activity of human ENaC, which results from a decrease in Na(+) self-inhibition. In the current work, we identified extracellular domain residues responsible for this regulation. We found that rat ENaC is less sensitive to pH than human ENaC, an effect mediated in part by the γ subunit. We identified a group of seven residues in the extracellular domain of γENaC (Asp-164, Gln-165, Asp-166, Glu-292, Asp-335, His-439, and Glu-455) that, when individually mutated to Ala, decreased proton activation of ENaC. γ(E455) is conserved in βENaC (Glu-446); mutation of this residue to neutral amino acids (Ala, Cys) reduced ENaC stimulation by acidic pH, whereas reintroduction of a negative charge (by MTSES modification of Cys) restored pH regulation. Combination of the seven γENaC mutations with β(E446A) generated a channel that was not activated by acidic pH, but inhibition by alkaline pH was intact. Moreover, these mutations reduced the effect of pH on Na(+) self-inhibition. Together, the data identify eight extracellular domain residues in human β- and γENaC that are required for regulation by acidic pH.","is_dataset_classified":null,"base_score":2.995732273553991,"endowment":2.995732273553991,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"23060445","pmcid":"PMC3510795","openalex_id":"https://openalex.org/W2036687784","authors":[],"funders":[{"funder_name":"NIDDK NIH HHS","grant_id":"DK25295","title":null},{"funder_name":"NHLBI NIH HHS","grant_id":"2T32HL007121-36","title":null},{"funder_name":"NHLBI NIH HHS","grant_id":"T32 HL007121","title":null}],"total_grants":3,"fwci":0.7367,"citation_percentile":0.68299324,"influential_citations":2,"citation_trend":[{"year":2014,"count":5},{"year":2015,"count":1},{"year":2016,"count":1},{"year":2018,"count":2},{"year":2019,"count":2},{"year":2022,"count":2},{"year":2023,"count":2},{"year":2024,"count":4}],"oa_status":"hybrid","license":"cc-by","oa_locations":[{"url":"http://www.jbc.org/article/S0021925820438955/pdf","host_type":"journal"},{"url":"http://www.jbc.org/article/S0021925820438955/pdf","host_type":"HYBRID"},{"url":"http://www.jbc.org/article/S0021925820438955/pdf","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S0021925820438955?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S0021925820438955?httpAccept=text/plain","host_type":"publisher"},{"url":"https://syndication.highwire.org/content/doi/10.1074/jbc.M112.417519","host_type":"publisher"},{"url":"https://doi.org/10.1074/jbc.m112.417519","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/23060445","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/3510795","host_type":"repository"}],"fields_of_study":["Ion Transport and Channel Regulation","Ion channel regulation and function","Neuroendocrine regulation and behavior","Biology","Medicine","Amino Acid Sequence","Animals","Biophysics","DNA, Complementary","Electrophysiology","Epithelial Sodium Channels","Female","Humans","Hydrogen-Ion Concentration","Hypertension","Kidney","Molecular Sequence Data","Mutagenesis, Site-Directed","Mutation","Oocytes","Protein Structure, Tertiary","Protons","Rats","Sequence Homology, Amino Acid","Sodium","Xenopus laevis"],"mesh_terms":["Amino Acid Sequence","Animals","Biophysics","Electrophysiology","Female","Humans","Hydrogen-Ion Concentration","Hypertension","Kidney","Molecular Sequence Data","Mutation","Oocytes","Protons","Sodium","Xenopus laevis","Mutagenesis, Site-Directed","Sequence Homology, Amino Acid","Protein Structure, Tertiary","DNA, Complementary","Rats","Epithelial Sodium Channels"],"keywords":["Epithelial sodium channel","Extracellular","Chemistry","Biochemistry","Protein subunit","Residue (chemistry)","Acid-sensing ion channel","Biophysics","Ion channel","Sodium","Biology","Receptor","Gene"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Clean water and sanitation"}],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[{"name":"pdb"}],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-07T07:11:53.184057Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
