Cloning of the epitope recognized by an OSM-neutralizing anti-gp130 monoclonal antibody identified a portion of gp130 receptor localized in the EF loop of the cytokine binding domain. Site-directed mutagenesis of the corresponding region was carried out by alanine substitution of residues 186-198. To generate type 1 or type 2 OSM receptors, gp130 mutants were expressed together with either LIF receptor beta or OSM receptor beta. When positions Val-189/Tyr-190 and Phe-191/Val-192 were alanine-substituted, Scatchard analyses indicated a complete abrogation of OSM binding to both type receptors. Interestingly, binding of LIF to type 1 receptor was not affected, corroborating the notion that in this case gp130 mostly behaves as a converter protein rather than a binding receptor. The present study demonstrates that positions 189-192 of gp130 cytokine binding domain are essential for OSM binding to both gp130/LIF receptor beta and gp130/OSM receptor beta heterocomplexes.","is_dataset_classified":null,"base_score":2.8903717578961645,"endowment":2.8903717578961645,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"10681548","pmcid":null,"openalex_id":"https://openalex.org/W2040724318","authors":[],"funders":[],"total_grants":0,"fwci":0.9394,"citation_percentile":0.75192712,"influential_citations":0,"citation_trend":[{"year":2013,"count":1},{"year":2015,"count":1},{"year":2022,"count":2},{"year":2023,"count":1},{"year":2024,"count":1}],"oa_status":"hybrid","license":"cc-by","oa_locations":[{"url":"http://www.jbc.org/article/S0021925818305945/pdf","host_type":"journal"},{"url":"http://www.jbc.org/article/S0021925818305945/pdf","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S0021925818305945?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S0021925818305945?httpAccept=text/plain","host_type":"publisher"},{"url":"https://syndication.highwire.org/content/doi/10.1074/jbc.275.8.5648","host_type":"publisher"},{"url":"https://doi.org/10.1074/jbc.275.8.5648","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/10681548","host_type":"repository"}],"fields_of_study":["Cytokine Signaling Pathways and Interactions","Monoclonal and Polyclonal Antibodies Research","Viral Infectious Diseases and Gene Expression in Insects"],"mesh_terms":["Alanine","Amino Acid Sequence","Animals","Antibodies, Monoclonal","Epitopes","Binding Sites","Dose-Response Relationship, Drug","Enzyme-Linked Immunosorbent Assay","Flow Cytometry","Growth Inhibitors","Humans","Kinetics","Lymphokines","Membrane Glycoproteins","Models, Molecular","Molecular Sequence Data","Peptides","Protein Binding","Recombinant Proteins","Antigens, CD","Interleukin-6","Cytokines","Mutagenesis, Site-Directed","Sequence Homology, Amino Acid","Protein Structure, Secondary","Receptors, Cytokine","Peptide Library","COS Cells","Cytokine Receptor gp130","Leukemia Inhibitory Factor","Receptors, OSM-LIF","Leukemia Inhibitory Factor Receptor alpha Subunit","Oncostatin M"],"keywords":["Oncostatin M","Glycoprotein 130","Interleukin-6 receptor","Leukemia inhibitory factor","Receptor","Leukemia inhibitory factor receptor","Interleukin-21 receptor","Cytokine receptor","Molecular biology","Interleukin-13 receptor","Biology","Ciliary neurotrophic factor","Chemistry","Cytokine","Biochemistry","Interleukin 6","Immunology","Insulin-like growth factor 1 receptor","Growth factor"],"sdg_mappings":[],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-04T14:29:59.714532Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
