Csk may be involved in the down-regulation of T cell receptor (TCR) signaling by C-terminal tyrosine phosphorylation of Lck and Fyn; however, it is not known how Csk activity is regulated or how it targets these Src family members. We used Jurkat T cells and normal human T cells to examine proteins that bind to the SH2 domain of Csk. In both Jurkat and normal T cells, the Src homology 2 (SH2) domain of Csk bound constitutively to a tyrosine-phosphorylated protein of 60 kDa (p60). The 60-kDa protein was detected in Csk immunoprecipitates from both unstimulated and CD3-stimulated cells. In addition to p60, a protein of 190 kDa coprecipitated with Csk, and both proteins were phosphorylated on tyrosine residues by the immunocomplex. Small amounts of GTPase-activating protein (GAP) were detected in anti-Csk immunoprecipitates, suggesting that p60 may be a GAP-associated protein. Our data demonstrate that the SH2 domain of Csk specifically associates with at least two tyrosine-phosphorylated proteins in normal human T cells, that this association is independent of TCR/CD3 activation, and that Csk may be a part of a multiprotein complex containing GAP.","is_dataset_classified":null,"base_score":2.1972245773362196,"endowment":2.1972245773362196,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"8621646","pmcid":null,"openalex_id":"https://openalex.org/W2069546498","authors":[],"funders":[{"funder_name":"NIAID NIH HHS","grant_id":"AI29575","title":null},{"funder_name":"NIAID NIH HHS","grant_id":"AI19727","title":null}],"total_grants":2,"fwci":0.3654,"citation_percentile":0.59286267,"influential_citations":0,"citation_trend":[],"oa_status":"hybrid","license":"cc-by","oa_locations":[{"url":"https://doi.org/10.1074/jbc.271.16.9698","host_type":"journal"},{"url":"https://doi.org/10.1074/jbc.271.16.9698","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S002192581731390X?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S002192581731390X?httpAccept=text/plain","host_type":"publisher"},{"url":"https://syndication.highwire.org/content/doi/10.1074/jbc.271.16.9698","host_type":"publisher"},{"url":"https://pubmed.ncbi.nlm.nih.gov/8621646","host_type":"repository"},{"url":"http://www.jbc.org/article/S002192581731390X/pdf","host_type":"Unpaywall"}],"fields_of_study":["T-cell and B-cell Immunology","Immune Cell Function and Interaction","Monoclonal and Polyclonal Antibodies Research","Amino Acid Sequence","CSK Tyrosine-Protein Kinase","Cell Line","Electrophoresis, Polyacrylamide Gel","Glutathione Transferase","Humans","Molecular Sequence Data","Molecular Weight","Phosphoproteins","Phosphotyrosine","Protein Binding","Protein-Tyrosine Kinases","Recombinant Fusion Proteins","T-Lymphocytes","Transfection","Tumor Cells, Cultured","src-Family Kinases"],"mesh_terms":["CSK Tyrosine-Protein Kinase","Amino Acid Sequence","Cell Line","Electrophoresis, Polyacrylamide Gel","Glutathione Transferase","Humans","Molecular Sequence Data","Molecular Weight","Phosphoproteins","Protein Binding","Protein-Tyrosine Kinases","Recombinant Fusion Proteins","T-Lymphocytes","Transfection","Tumor Cells, Cultured","Phosphotyrosine","src-Family Kinases"],"keywords":["Tyrosine-protein kinase CSK","SH2 domain","FYN","Proto-oncogene tyrosine-protein kinase Src","SH3 domain","Tyrosine phosphorylation","Jurkat cells","Tyrosine kinase","Cell biology","Receptor tyrosine kinase","Biology","Protein tyrosine phosphatase","Tyrosine","GTPase-activating protein","Phosphorylation","Chemistry","Biochemistry","Signal transduction","T cell","G protein","Immunology"],"sdg_mappings":[],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-18T18:30:43.072445Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
