This family contains the BLM and WRN helicases, in which germline mutations are responsible for Bloom and Werner syndromes, respectively. TFIIH DNA helicases, XPB and XPD, are also components in this apoptotic pathway. We hypothesized that there may be some redundancy between helicases in their ability to complement the attenuated p53-mediated apoptotic levels seen in cells from individuals with diseases associated with these defective helicase genes. The attenuated apoptotic phenotype in Bloom syndrome cells was rescued not only by ectopic expression of BLM, but also by WRN or XPB, both 3' --> 5' helicases, but not expression of the 5' --> 3' helicase XPD. Overexpression of Sgs1, a WRN/BLM yeast homolog, corrected the reduction in BS cells only, which is consistent with Sgs1 being evolutionarily most homologous to BLM. A restoration of apoptotic levels in cells from WS, XPB or XPD patients was attained only by overexpression of the specific helicase. Our data suggest a limited redundancy in the pathways of these RecQ helicases in p53-induced apoptosis.","is_dataset_classified":null,"base_score":3.332204510175204,"endowment":3.332204510175204,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"16288211","pmcid":"PMC1420682","openalex_id":"https://openalex.org/W2017592458","authors":[],"funders":[{"funder_name":"NCI NIH HHS","grant_id":"Z01 BC005795-11","title":null},{"funder_name":"Intramural NIH HHS","grant_id":"","title":null}],"total_grants":2,"fwci":1.661,"citation_percentile":0.82176932,"influential_citations":0,"citation_trend":[{"year":2012,"count":3},{"year":2013,"count":1},{"year":2014,"count":1},{"year":2019,"count":2},{"year":2024,"count":2}],"oa_status":"green","license":"https://www.springer.com/tdm","oa_locations":[{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/1420682","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/1420682","host_type":"repository"},{"url":"https://www.nature.com/articles/1209242.pdf","host_type":"publisher"},{"url":"https://www.nature.com/articles/1209242","host_type":"publisher"},{"url":"https://doi.org/10.1038/sj.onc.1209242","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/16288211","host_type":"repository"}],"fields_of_study":["DNA Repair Mechanisms","Cancer-related Molecular Pathways","Carcinogens and Genotoxicity Assessment"],"mesh_terms":["Bloom Syndrome","DNA Helicases","Humans","Werner Syndrome","Tumor Suppressor Protein p53","Apoptosis","Germ-Line Mutation"],"keywords":["Helicase","Bloom syndrome","Biology","DNA repair","Genome instability","Werner syndrome","Genetics","Apoptosis","Cell biology","Molecular biology","DNA damage","Gene","DNA","RNA"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Life in Land"}],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[{"name":"igsr"}],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-16T16:45:50.218130Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
