Hypoxia stimulates the secretion of vascular endothelial growth factor (VEGF) and other angiogenic factors, leading to neovascularization and protection against ischaemic injury. Here we show that the transcriptional coactivator PGC-1alpha (peroxisome-proliferator-activated receptor-gamma coactivator-1alpha), a potent metabolic sensor and regulator, is induced by a lack of nutrients and oxygen, and PGC-1alpha powerfully regulates VEGF expression and angiogenesis in cultured muscle cells and skeletal muscle in vivo. PGC-1alpha-/- mice show a striking failure to reconstitute blood flow in a normal manner to the limb after an ischaemic insult, whereas transgenic expression of PGC-1alpha in skeletal muscle is protective. Surprisingly, the induction of VEGF by PGC-1alpha does not involve the canonical hypoxia response pathway and hypoxia inducible factor (HIF). Instead, PGC-1alpha coactivates the orphan nuclear receptor ERR-alpha (oestrogen-related receptor-alpha) on conserved binding sites found in the promoter and in a cluster within the first intron of the VEGF gene. Thus, PGC-1alpha and ERR-alpha, major regulators of mitochondrial function in response to exercise and other stimuli, also control a novel angiogenic pathway that delivers needed oxygen and substrates. PGC-1alpha may provide a novel therapeutic target for treating ischaemic diseases.","is_dataset_classified":null,"base_score":6.960347729101308,"endowment":6.960347729101308,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"18288196","pmcid":null,"openalex_id":"https://openalex.org/W2083456009","authors":[],"funders":[{"funder_name":"NIDDK NIH HHS","grant_id":"P30 DK040561","title":null},{"funder_name":"NIDDK NIH HHS","grant_id":"R01 DK054477","title":null}],"total_grants":2,"fwci":34.0072,"citation_percentile":0.99909634,"influential_citations":0,"citation_trend":[{"year":2012,"count":90},{"year":2013,"count":77},{"year":2014,"count":91},{"year":2015,"count":66},{"year":2016,"count":63},{"year":2017,"count":58},{"year":2018,"count":59},{"year":2019,"count":44},{"year":2020,"count":53},{"year":2021,"count":53},{"year":2022,"count":61},{"year":2023,"count":42},{"year":2024,"count":45},{"year":2025,"count":36},{"year":2026,"count":10}],"oa_status":"bronze","license":"http://www.springer.com/tdm","oa_locations":[{"url":"https://www.nature.com/articles/nature06613.pdf","host_type":"journal"},{"url":"https://www.nature.com/articles/nature06613.pdf","host_type":"publisher"},{"url":"http://www.nature.com/articles/nature06613","host_type":"publisher"},{"url":"http://www.nature.com/articles/nature06613.pdf","host_type":"publisher"},{"url":"https://doi.org/10.1038/nature06613","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/18288196","host_type":"repository"},{"url":"https://www.nature.com/articles/nature06613","host_type":"repository"}],"fields_of_study":["Adipose Tissue and Metabolism","Cancer, Hypoxia, and Metabolism","Peroxisome Proliferator-Activated Receptors"],"mesh_terms":["Peroxisome Proliferator-Activated Receptor Gamma Coactivator 1-alpha","ERRalpha Estrogen-Related Receptor","Animals","Cells, Cultured","Gene Expression Regulation","Ischemia","Mice, Transgenic","Oxygen","Receptors, Estrogen","Transcription Factors","Trans-Activators","Cell Hypoxia","Muscle, Skeletal","Neovascularization, Physiologic","Transgenes","Vascular Endothelial Growth Factor A","Mice","Hypoxia-Inducible Factor 1"],"keywords":["Angiogenesis","Coactivator","Regulator","Mitochondrial biogenesis","Hypoxia (environmental)","HIF1A","Biology","Vascular endothelial growth factor","Skeletal muscle","Endocrinology","Neovascularization","Hypoxia-inducible factors","Internal medicine","Cell biology","Transcription factor","Cancer research","Mitochondrion","Chemistry","Medicine","Biochemistry","VEGF receptors"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Good health and well-being"}],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-07-31T07:33:19.433256Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
