Germinal centers (GCs) in Peyer's patches (PPs) are the principal inductive sites where naive B cells specific for bacterial antigens encounter their cognate antigens and receive T-cell help driving their differentiation into IgA-producing plasma cells. We investigated the role of antigen sampling by intestinal M cells in initiating the SIgA response to gut bacteria by developing mice in which receptor activator of nuclear factor-κB ligand (RANKL)-dependent M-cell differentiation was abrogated by conditional deletion of Tnfrsf11a in the intestinal epithelium. Mice without intestinal M cells had profound delays in PP GC maturation and emergence of lamina propria IgA plasma cells, resulting in diminished levels of fecal SIgA that persisted into adulthood. We conclude that M-cell-mediated sampling of commensal bacteria is a required initial step for the efficient induction of intestinal SIgA.","is_dataset_classified":null,"base_score":5.583496308781699,"endowment":5.583496308781699,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"26601902","pmcid":"PMC4917673","openalex_id":"https://openalex.org/W2174345167","authors":[],"funders":[{"funder_name":"NIDDK NIH HHS","grant_id":"R24 DK064399","title":null},{"funder_name":"NIAID NIH HHS","grant_id":"R21 AI111388","title":null},{"funder_name":"NIDDK NIH HHS","grant_id":"R01 DK064730","title":null},{"funder_name":"National Institutes of Health","grant_id":"5R21AI111388-02","title":"Intestinal M Cells and Secretory IgA Response to Defined Gut Microbiota"},{"funder_name":"National Institutes of Health","grant_id":"5R01DK064730-03","title":"CCR6 Regulation of Intestinal T Lymphocyte Development"}],"total_grants":5,"fwci":6.0581,"citation_percentile":0.97198021,"influential_citations":0,"citation_trend":[{"year":2015,"count":1},{"year":2016,"count":9},{"year":2017,"count":19},{"year":2018,"count":19},{"year":2019,"count":33},{"year":2020,"count":32},{"year":2021,"count":33},{"year":2022,"count":30},{"year":2023,"count":28},{"year":2024,"count":30},{"year":2025,"count":14},{"year":2026,"count":17}],"oa_status":"hybrid","license":"cc-by-nc-sa","oa_locations":[{"url":"https://www.nature.com/articles/mi2015121.pdf","host_type":"journal"},{"url":"https://www.nature.com/articles/mi2015121.pdf","host_type":"publisher"},{"url":"https://www.nature.com/articles/mi2015121","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S1933021922009151?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S1933021922009151?httpAccept=text/plain","host_type":"publisher"},{"url":"https://doi.org/10.1038/mi.2015.121","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/26601902","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/4917673","host_type":"repository"},{"url":"https://europepmc.org/articles/PMC4917673","host_type":"Europe_PMC"},{"url":"https://europepmc.org/articles/PMC4917673?pdf=render","host_type":"Europe_PMC"},{"url":"http://dx.doi.org/10.1038/mi.2015.121","host_type":""},{"url":"https://dx.doi.org/10.1038/mi.2015.121","host_type":""}],"fields_of_study":["Immune Cell Function and Interaction","T-cell and B-cell Immunology","Immunotherapy and Immune Responses","0301 basic medicine","03 medical and health sciences","0302 clinical medicine","Animals","Antigen Presentation","Antigens, Bacterial","B-Lymphocytes","Cell Differentiation","Cells, Cultured","Gastrointestinal Microbiome","Germinal Center","Homeostasis","Immunity, Humoral","Immunity, Mucosal","Immunoglobulin A, Secretory","Intestinal Mucosa","Mice","Mice, Inbred C57BL","Mice, Knockout","Peyer's Patches","Receptor Activator of Nuclear Factor-kappa B","Symbiosis","T-Lymphocytes, Helper-Inducer"],"mesh_terms":["Gastrointestinal Microbiome","Animals","Antigens, Bacterial","B-Lymphocytes","Cell Differentiation","Cells, Cultured","T-Lymphocytes, Helper-Inducer","Homeostasis","Immunoglobulin A, Secretory","Intestinal Mucosa","Mice, Inbred C57BL","Peyer's Patches","Symbiosis","Antigen Presentation","Mice, Knockout","Germinal Center","Immunity, Mucosal","Mice","Receptor Activator of Nuclear Factor-kappa B","Immunity, Humoral"],"keywords":["Microfold cell","Biology","Antigen","Immunology","Immune system","Lamina propria","Mucosal immunology","Microbiology","Germinal center","Immunoglobulin A","B cell","Epithelium","Antibody","Immunity","Immunoglobulin G","Article","Mice","Peyer's Patches","Animals","Homeostasis","Intestinal Mucosa","Symbiosis","Immunity, Mucosal","Cells, Cultured","Mice, Knockout","Antigen Presentation","Antigens, Bacterial","B-Lymphocytes","Receptor Activator of Nuclear Factor-kappa B","Cell Differentiation","T-Lymphocytes, Helper-Inducer","Gastrointestinal Microbiome","Immunity, Humoral","Mice, Inbred C57BL","Immunoglobulin A, Secretory"],"sdg_mappings":[{"sdg_number":3,"sdg_label":"3.
