Eleven alterations (one in exon 1, five in exon 2 and five in the 3' non-coding sequence of the exon 3 region) were concordantly and independently detected by both SSCP and nucleotide sequence analysis. Two of the exon 2 changes and the five changes in the non-coding exon 3 region are likely to represent natural polymorphism. Four (15%) of 26 metastases thus had CDKN2 mutations and belonged to 3 (12%) of 25 patients. Semi-quantitative PCR furthermore revealed no sign of homozygous deletions of the CDKN2 exon 2 region. The results support an involvement of the CDKN2 product in the development of a subgroup of sporadic melanomas and encourage the search for alterations in additional genes of the 9p21 region.","is_dataset_classified":null,"base_score":3.5263605246161616,"endowment":3.5263605246161616,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"8562340","pmcid":"PMC2074427","openalex_id":"https://openalex.org/W2030119528","authors":[],"funders":[],"total_grants":0,"fwci":2.8343,"citation_percentile":0.91796109,"influential_citations":0,"citation_trend":[],"oa_status":"bronze","license":"cc-by","oa_locations":[{"url":"https://www.nature.com/articles/bjc199659.pdf","host_type":"journal"},{"url":"https://www.nature.com/articles/bjc199659.pdf","host_type":"publisher"},{"url":"http://www.nature.com/articles/bjc199659.pdf","host_type":"publisher"},{"url":"http://www.nature.com/articles/bjc199659","host_type":"publisher"},{"url":"https://doi.org/10.1038/bjc.1996.59","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/8562340","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/2074427","host_type":"repository"},{"url":"https://europepmc.org/articles/PMC2074427","host_type":"Europe_PMC"},{"url":"https://europepmc.org/articles/PMC2074427?pdf=render","host_type":"Europe_PMC"}],"fields_of_study":["Cancer Genomics and Diagnostics","Cutaneous Melanoma Detection and Management","Immunotherapy and Immune Responses","Alleles","Amino Acid Sequence","Base Sequence","Carrier Proteins","Cyclin-Dependent Kinase Inhibitor p16","DNA Primers","DNA, Neoplasm","Female","Genes, Tumor Suppressor","Humans","Male","Melanoma","Molecular Sequence Data","Neoplasm Metastasis","Polymorphism, Single-Stranded Conformational","Protein Kinase Inhibitors","Sequence Deletion","Skin Neoplasms"],"mesh_terms":["Alleles","Amino Acid Sequence","Base Sequence","Carrier Proteins","DNA, Neoplasm","Female","Humans","Male","Melanoma","Molecular Sequence Data","Neoplasm Metastasis","Skin Neoplasms","Genes, Tumor Suppressor","Sequence Deletion","DNA Primers","Polymorphism, Single-Stranded Conformational","Cyclin-Dependent Kinase Inhibitor p16","Protein Kinase Inhibitors"],"keywords":["Exon","Single-strand conformation polymorphism","Coding region","Biology","Polymerase chain reaction","Gene","Genetics","DNA sequencing","Melanoma","Sequence analysis","Molecular biology","Cancer research"],"sdg_mappings":[],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-03T14:50:42.542206Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
