{"doi":"10.1021/acschembio.0c00196","title":"A Single Reactive Noncanonical Amino Acid is Able to Dramatically Stabilize Protein Structure","abstract":"A p-isothiocyanate phenylalanine\\nmutant of the homodimeric protein\\nhomoserine o-succinyltransferase (MetA) was isolated in a temperature\\ndependent selection from a library of <i>metA</i> mutants\\ncontaining noncanonical amino acids. This mutant protein has a dramatic\\nincrease of 24 °C in thermal stability compared to the wild type\\nprotein. Peptide mapping experiments revealed that the isothiocyanate\\ngroup forms a thiourea cross-link to the N terminal proline of the\\nother monomer, despite the two positions being >30 Å apart\\nin\\nthe X-ray crystal structure of the wild type protein.
