This study analyzed a diverse LUAD patient cohort with EGFR mutation statuses and wild-type profiles for ALK and KRAS to identify stage-specific biomarkers. Using quantitative proteomics and multiomics, we discovered 21 dysregulated proteins in early-stage EGFR-WT LUAD, identifying myeloid-derived growth factor (MYDGF) as a key candidate biomarker. Elevated MYDGF levels in tissue ( n = 117) and serum ( n = 196) correlated significantly with cancer stage in EGFR-WT patients but not EGFR-MT cases. Notably, a higher tumor-to-normal MYDGF ratio predicted a favorable prognosis in early-stage EGFR-WT LUAD. Functional studies demonstrated that MYDGF exerts distinct roles in cell viability and migration depending on its cellular localization and the invasive potential of cancer cells. Specifically, secreted MYDGF promoted a protumorigenic phenotype, whereas excess intracellular MYDGF appeared to suppress the oncogenic capacity of aggressive cancer cells. MYDGF knockdown and subsequent proteomic analysis provided further insights into these context-dependent functions. These findings highlight EGFR status- and stage-specific proteomic profiles in LUAD, emphasizing the importance of context-dependent biomarker assessment for personalized treatment strategies.","is_dataset_classified":null,"base_score":1.6094379124341003,"endowment":1.6094379124341003,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"40752010","pmcid":null,"openalex_id":"https://openalex.org/W4412862175","authors":[],"funders":[{"funder_name":"Chang Gung Memorial Hospital","grant_id":"BMRP894","title":null},{"funder_name":"Chang Gung Memorial Hospital","grant_id":"CMRPD1M0441-3","title":null},{"funder_name":"National Science and Technology Council","grant_id":"108-2320-B-182-007-MY3","title":null},{"funder_name":"National Science and Technology Council","grant_id":"111-2320-B-182-019-MY3","title":null},{"funder_name":"Chang Gung University","grant_id":"","title":null},{"funder_name":"Ministry of Education, Taiwan","grant_id":"","title":null}],"total_grants":6,"fwci":3.0225,"citation_percentile":0.91786697,"influential_citations":0,"citation_trend":[{"year":2025,"count":1},{"year":2026,"count":3}],"oa_status":"green","license":"https://creativecommons.org/licenses/by-nc-nd/4.0/","oa_locations":[{"url":"https://pmc.ncbi.nlm.nih.gov/articles/PMC12419241/","host_type":"repository"},{"url":"https://pmc.ncbi.nlm.nih.gov/articles/PMC12419241/","host_type":"repository"},{"url":"https://pubs.acs.org/doi/pdf/10.1021/acs.jproteome.5c00385","host_type":"publisher"},{"url":"https://doi.org/10.1021/acs.jproteome.5c00385","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/40752010","host_type":"repository"},{"url":"http://ir.nhri.org.tw/handle/3990099045/17302","host_type":"repository"}],"fields_of_study":["Lung Cancer Treatments and Mutations","Lung Cancer Research Studies","Peptidase Inhibition and Analysis","Medicine","Biology","Humans","ErbB Receptors","Adenocarcinoma of Lung","Lung Neoplasms","Biomarkers, Tumor","Female","Proteomics","Male","Mutation","Carcinogenesis","Cell Line, Tumor","Middle Aged","Cell Movement","Prognosis","Gene Expression Regulation, Neoplastic","Aged"],"mesh_terms":["Adenocarcinoma of Lung","Aged","Cell Movement","Female","Humans","Lung Neoplasms","Male","Middle Aged","Mutation","Prognosis","Biomarkers, Tumor","Gene Expression Regulation, Neoplastic","Proteomics","Cell Line, Tumor","Carcinogenesis","ErbB Receptors"],"keywords":["Carcinogenesis","Lung cancer","Cancer research","Adenocarcinoma","Oncology","Cancer","Epidermal growth factor receptor","Biology","Internal medicine","Medicine","Biomarker","Lung adenocarcinoma","Quantitative Proteomics","Wild-type Egfr","Myeloid-derived Growth Factor"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Good health and well-being"}],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-07-31T17:57:18.692926Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
