Detection of this digestion and appetite-regulating hormone currently relies on the use of immunoassays, many of which suffer from insufficient sensitivity in the physiological range and cross-reactivity problems with gastrin, which circulates at higher plasma concentrations. As an alternative to existing techniques, a liquid chromatography and mass spectrometry-based method was developed to measure CCK-derived peptides in cell culture supernatants. The method was initially applied to organoid studies and was capable of detecting both CCK8 and an N-terminal peptide fragment (prepro) ppCCK(21-44) in supernatants following stimulation. Extraction optimization was performed using statistical modeling software, enabling a quantitative LC-MS/MS method for ppCCK(21-44) capable of detecting this peptide in the low pM range in human plasma and secretion buffer solutions. Plasma samples from healthy individuals receiving a standardized meal (Ensure) after an overnight fast were analyzed; however, the method only had sensitivity to detect ppCCK(21-44). Secretion studies employing human intestinal organoids and meal studies in healthy volunteers confirmed that ppCCK(21-44) is a suitable surrogate analyte for measuring the release of CCK in vitro and in vivo.","is_dataset_classified":null,"base_score":2.1972245773362196,"endowment":2.1972245773362196,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"37591880","pmcid":"PMC10476265","openalex_id":"https://openalex.org/W4385969291","authors":[],"funders":[{"funder_name":"Medical Research Council","grant_id":"MC_UU_00014/3","title":"Entero-endocrinology including the effects of bariatric surgery"},{"funder_name":"Medical Research Council","grant_id":"MR/M009041/1","title":"Innovative Technologies for Stratified and Experimental Medicine"},{"funder_name":"Medical Research Council","grant_id":"MRC_MC_UU_00014/5","title":null},{"funder_name":"Wellcome Trust","grant_id":"100574/Z/12/Z","title":null},{"funder_name":"Wellcome Trust","grant_id":"220271/Z/20/Z","title":null},{"funder_name":"Medical Research Council","grant_id":"MC_UU_12012/1","title":null},{"funder_name":"Medical Research Council","grant_id":"MC_UU_00014/5","title":null},{"funder_name":"Medical Research Council","grant_id":"MR/X012417/1","title":null},{"funder_name":"Wellcome Trust","grant_id":"220271","title":"Targeting the gut in metabolic disease"},{"funder_name":"Wellcome Trust","grant_id":"100574","title":"Institute of Metabolic Science."},{"funder_name":"University of Cambridge","grant_id":"","title":null},{"funder_name":"Wellcome Trust","grant_id":"","title":null},{"funder_name":"National Institute for Health Research (NIHR)","grant_id":"","title":null},{"funder_name":"Biotechnology and Biological Sciences Research Council","grant_id":"","title":null},{"funder_name":"Wellcome Trust","grant_id":"","title":null},{"funder_name":"Department of Health","grant_id":"","title":null},{"funder_name":"Biotechnology and Biological Sciences Research Council","grant_id":"","title":null}],"total_grants":17,"fwci":1.0444,"citation_percentile":0.73668374,"influential_citations":0,"citation_trend":[{"year":2024,"count":3},{"year":2025,"count":5}],"oa_status":"hybrid","license":"cc-by","oa_locations":[{"url":"https://pubs.acs.org/doi/pdf/10.1021/acs.jproteome.3c00272","host_type":"journal"},{"url":"https://pubs.acs.org/doi/pdf/10.1021/acs.jproteome.3c00272","host_type":"publisher"},{"url":"https://doi.org/10.1021/acs.jproteome.3c00272","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/37591880","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/10476265","host_type":"repository"},{"url":"https://www.repository.cam.ac.uk/handle/1810/356885","host_type":"repository"},{"url":"https://pmc.ncbi.nlm.nih.gov/articles/PMC10476265/pdf/pr3c00272.pdf","host_type":"repository"},{"url":"https://www.repository.cam.ac.uk/bitstreams/41273793-b2a6-4f42-8a32-9a01b9392115/download","host_type":"repository"},{"url":"https://europepmc.org/articles/PMC10476265","host_type":"Europe_PMC"},{"url":"https://europepmc.org/articles/PMC10476265?pdf=render","host_type":"Europe_PMC"},{"url":"https://dx.doi.org/10.17863/cam.100110","host_type":""},{"url":"http://dx.doi.org/10.1021/acs.jproteome.3c00272","host_type":""},{"url":"https://www.repository.cam.ac.uk/handle/1810/354242","host_type":""},{"url":"https://doi.org/10.17863/cam.100110","host_type":""}],"fields_of_study":["Neuropeptides and Animal Physiology","Receptor Mechanisms and Signaling","Estrogen and related hormone effects","0303 health sciences","03 medical and health sciences","Humans","Cholecystokinin","Chromatography, Liquid","Tandem Mass Spectrometry","Biological Transport","Bodily Secretions"],"mesh_terms":["Biological Transport","Cholecystokinin","Chromatography, Liquid","Humans","Bodily Secretions","Tandem Mass Spectrometry"],"keywords":["Cholecystokinin","Enteroendocrine cell","Analyte","Chromatography","Gastrin","Chemistry","In vivo","Peptide","Secretion","Hormone","Biology","Biochemistry","Endocrine system","Experimental design","Liquid chromatography-mass spectrometry","Bodily Secretions","Tandem Mass Spectrometry","Humans","Biological Transport","Chromatography, Liquid"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Good health and well-being"}],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-07-29T18:08:52.907043Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
