More than two centuries ago, autopsy pathologists noted an increased prevalence of cirrhosis in alcoholic subjects. 28 Later, population-based studies identified a correlation between liver disease mortality and per capita consumption of alcoholic beverages. 12 More recently, carefully-controlled studies in well-nourished animal models have clearly demonstrated that both the prevalence and the severity of liver disease increase with chronic consumption of alcohol. 31 Current efforts focus on explaining interindividual variability in susceptibility to alcohol-mediated liver injury. Success in the latter endeavor will require a more detailed understanding of the mechanisms involved in the pathogenesis of alcoholic liver disease (ALD). Such knowledge should, in turn, suggest strategies to prevent and treat ALD.
