It is an immediate-early gene, regulated at many levels of its biosynthetic pathway. Herein we show that the serine/threonine protein kinase CK1 phosphorylates RhoB in vitro but not RhoA or RhoC. With the use of specific CK1 inhibitors, IC261 and D4476, we show that the kinase phosphorylates also RhoB in HeLa cells. Mass spectrometry analysis demonstrates that RhoB is monophosphorylated by CK1, in its C-terminal end, on serine 185. The substitution of Ser185 by Ala dramatically inhibited the phosphorylation of RhoB in cultured cells. Lastly we show that the inhibition of CK1 activates RhoB and promotes RhoB dependent actin fiber formation and EGF-R level. Our data provide the first demonstration of RhoB phosphorylation and indicate that this post-translational maturation would be a novel critical mechanism to control the RhoB functions.","is_dataset_classified":null,"base_score":3.332204510175204,"endowment":3.332204510175204,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"18590726","pmcid":null,"openalex_id":"https://openalex.org/W2028352012","authors":[],"funders":[{"funder_name":"Engineering and Physical Sciences Research Council","grant_id":"EP/C007247/1","title":null}],"total_grants":1,"fwci":1.1698,"citation_percentile":0.76722734,"influential_citations":0,"citation_trend":[{"year":2012,"count":4},{"year":2013,"count":1},{"year":2014,"count":3},{"year":2015,"count":1},{"year":2016,"count":3},{"year":2020,"count":2},{"year":2021,"count":1},{"year":2022,"count":1},{"year":2023,"count":1},{"year":2024,"count":1}],"oa_status":"closed","license":"https://doi.org/10.15223/policy-004","oa_locations":[{"url":"https://api.elsevier.com/content/article/PII:S0014482708002425?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S0014482708002425?httpAccept=text/plain","host_type":"publisher"},{"url":"https://doi.org/10.1016/j.yexcr.2008.06.011","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/18590726","host_type":"repository"},{"url":"http://eprints.gla.ac.uk/44906/","host_type":"journal"}],"fields_of_study":["Protein Kinase Regulation and GTPase Signaling","Cellular Mechanics and Interactions","Melanoma and MAPK Pathways","Actin Cytoskeleton","Actins","Amino Acid Sequence","Amino Acid Substitution","Casein Kinase Ialpha","Enzyme Activation","Enzyme Inhibitors","ErbB Receptors","HeLa Cells","Humans","Molecular Sequence Data","Phosphorylation","Protein Processing, Post-Translational","Protein Serine-Threonine Kinases","Sequence Homology, Amino Acid","Serine","Stress Fibers","rhoB GTP-Binding Protein"],"mesh_terms":["Actins","Amino Acid Sequence","Enzyme Activation","Enzyme Inhibitors","HeLa Cells","Humans","Actin Cytoskeleton","Molecular Sequence Data","Phosphorylation","Protein Processing, Post-Translational","Serine","Protein Serine-Threonine Kinases","Sequence Homology, Amino Acid","Amino Acid Substitution","rhoB GTP-Binding Protein","Stress Fibers","Casein Kinase Ialpha","ErbB Receptors","Hela Cells"],"keywords":["RHOB","RHOA","Biology","Phosphorylation","Stress fiber","RhoC","Cell biology","Serine","Signal transduction","Focal adhesion"],"sdg_mappings":[],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-07-29T14:58:18.353425Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
