Given the risk of a potential IAV pandemic, it is increasingly important to better understand virus-host interactions and develop new anti-viral strategies. Here, we reported nonmuscle myosin IIA (MYH9)-mediated regulation of IAV infection. MYH9 depletion caused a profound inhibition of IAV infection by reducing viral attachment and internalization in human lung epithelial cells. Surprisingly, overexpression of MYH9 also led to a significant reduction in viral productive infection. Interestingly, overexpression of MYH9 retained viral attachment, internalization, or uncoating, but suppressed the viral ribonucleoprotein (vRNP) activity in a minigenome system. Further analyses found that excess MYH9 might interrupt the formation of vRNP by interacting with the viral nucleoprotein (NP) and result in the reduction of the completed vRNP in the nucleus, thereby inhibiting subsequent viral RNA transcription and replication. Together, we discovered that MYH9 can interact with IAV NP protein and engage in the regulation of vRNP complexes, thereby involving viral replication. These findings enlighten new mechanistic insights into the complicated interface of host-IAV interactions, ultimately making it an attractive target for the generation of antiviral drugs.","is_dataset_classified":null,"base_score":2.1972245773362196,"endowment":2.1972245773362196,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"36509386","pmcid":"PMC10006312","openalex_id":"https://openalex.org/W4311141316","authors":[],"funders":[{"funder_name":"National Natural Science Foundation of China","grant_id":"81771704","title":null},{"funder_name":"National Natural Science Foundation of China","grant_id":"81901598","title":null},{"funder_name":"National Natural Science Foundation of China","grant_id":"82041015","title":null},{"funder_name":"National Natural Science Foundation of China","grant_id":"82071788","title":null},{"funder_name":"National Key Research and Development Program of China","grant_id":"2022YFC2604100","title":null}],"total_grants":5,"fwci":1.1739,"citation_percentile":0.78536699,"influential_citations":0,"citation_trend":[{"year":2023,"count":1},{"year":2024,"count":2},{"year":2025,"count":5}],"oa_status":"hybrid","license":"cc-by-nc-nd","oa_locations":[{"url":"https://www.sciencedirect.com/science/article/pii/S1995820X22002085/pdf","host_type":"journal"},{"url":"https://www.sciencedirect.com/science/article/pii/S1995820X22002085/pdf","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S1995820X22002085?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S1995820X22002085?httpAccept=text/plain","host_type":"publisher"},{"url":"https://doi.org/10.1016/j.virs.2022.12.002","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/36509386","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/10006312","host_type":"repository"},{"url":"https://pmc.ncbi.nlm.nih.gov/articles/PMC10006312/pdf/main.pdf","host_type":"repository"},{"url":"https://europepmc.org/articles/PMC10006312","host_type":"Europe_PMC"},{"url":"https://europepmc.org/articles/PMC10006312?pdf=render","host_type":"Europe_PMC"}],"fields_of_study":["Influenza Virus Research Studies","RNA modifications and cancer","Respiratory viral infections research","Humans","Host-Pathogen Interactions","Influenza A virus","Influenza, Human","Lung","Nonmuscle Myosin Type IIA","Nucleoproteins","Nucleotidyltransferases","Virus Internalization","Virus Replication","Myosin Heavy Chains"],"mesh_terms":["Humans","Influenza, Human","Lung","Nucleoproteins","Nucleotidyltransferases","Influenza A virus","Virus Replication","Myosin Heavy Chains","Nonmuscle Myosin Type IIA","Virus Internalization","Host-Pathogen Interactions"],"keywords":["Nucleoprotein","Ribonucleoprotein","Internalization","Influenza A virus","Virology","Virus","Viral replication","Viral matrix protein","Biology","Cell biology","RNA","Cell","Gene","Genetics","virus entry","Virus-host Interactions","Vrnp Activity","Influenza A Virus (Iav)","Myosin Iia (Myh9)"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Good health and well-being"}],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[{"name":"gen"},{"name":"uniprot"}],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-06T18:28:29.786056Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
