An issue that has remained controversial is whether the editing of deoxycytidine residues to deoxyuridine is necessary and sufficient for this inhibition or whether APOBEC3 proteins also exert a second, distinct inhibitory mechanism. Here, we present an analysis of the ability of mutants of APOBEC3G and APOBEC3B, both of which contain two consensus cytidine deaminase active sites, to inhibit the replication of human immunodeficiency virus. Our data confirm that APOBEC3G only contains a single, carboxy-terminal active site but, surprisingly, reveal that both cytidine deaminase consensus sequences in APOBEC3B are enzymatically active. Enzymatically inactive mutant forms of APOBEC3G and APOBEC3B were found to retain the ability to inhibit the infectivity of HIV-1 virions produced in their presence by approximately 4-fold and approximately 8-fold, respectively. While this inhibition was significantly less than the level seen with wild-type forms of A3G or A3B, these data, nevertheless argue that the inhibition of HIV-1 by APOBEC3 proteins is at least partly independent of DNA editing.","is_dataset_classified":null,"base_score":4.248495242049359,"endowment":4.248495242049359,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"17434555","pmcid":"PMC1950718","openalex_id":"https://openalex.org/W2041401655","authors":[],"funders":[{"funder_name":"National Institutes of Health","grant_id":"AI057099","title":null},{"funder_name":"National Institutes of Health","grant_id":"AI064518","title":null},{"funder_name":"National Institutes of Health","grant_id":"AI065301","title":null},{"funder_name":"NIAID NIH HHS","grant_id":"P30 AI064518","title":null},{"funder_name":"NIAID NIH HHS","grant_id":"R01 AI057099","title":null},{"funder_name":"NIAID NIH HHS","grant_id":"R01 AI065301","title":null}],"total_grants":6,"fwci":2.8747,"citation_percentile":0.91314896,"influential_citations":0,"citation_trend":[{"year":2012,"count":7},{"year":2013,"count":4},{"year":2014,"count":3},{"year":2015,"count":4},{"year":2016,"count":3},{"year":2017,"count":6},{"year":2018,"count":1},{"year":2019,"count":4},{"year":2021,"count":2},{"year":2022,"count":1},{"year":2025,"count":1}],"oa_status":"bronze","license":"http://www.elsevier.com/open-access/userlicense/1.0/","oa_locations":[{"url":"https://www.sciencedirect.com/science/article/pii/S0042682207001778/pdf","host_type":"journal"},{"url":"https://www.sciencedirect.com/science/article/pii/S0042682207001778/pdf","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S0042682207001778?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S0042682207001778?httpAccept=text/plain","host_type":"publisher"},{"url":"https://doi.org/10.1016/j.virol.2007.03.019","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/17434555","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/1950718","host_type":"repository"}],"fields_of_study":["HIV Research and Treatment","HIV/AIDS drug development and treatment","Cytomegalovirus and herpesvirus research"],"mesh_terms":["APOBEC-3G Deaminase","Cell Line","Cytidine Deaminase","Humans","Nucleoside Deaminases","Repressor Proteins","RNA, Viral","HIV-1","Minor Histocompatibility Antigens","Mutagenesis, Site-Directed","RNA Editing","Protein Structure, Tertiary","Amino Acid Substitution","Anti-Retroviral Agents","In Vitro Techniques"],"keywords":["Cytidine deaminase","APOBEC3G","Cytidine","Biology","Activation-induced (cytidine) deaminase","Mutant","Retrovirus","Biochemistry","Viral replication","Enzyme","Molecular biology","Cell biology","Virology","Genetics","Virus","Gene","Somatic hypermutation","Antibody"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Good health and well-being"}],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-03T03:57:22.455801Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
