It remains unclear, however, whether functionally selective properties are meaningful in vivo. Data obtained with experimental compounds that are functionally selective at the dopamine D2L receptor in vitro suggest that these properties might predict atypical behavioral actions. Moreover, the antipsychotic drug aripiprazole is commonly thought to be a D2 partial agonist, but data clearly show that aripiprazole is functionally selective in vitro. It is proposed that the effects of aripiprazole in animal models and humans can be reconciled only with its functionally selective D2 properties, not its partial D2 agonism. Together, these data provide support for the hypothesis that compounds with functionally selective properties in vitro are likely to have novel actions in vivo, opening doors to new avenues of drug discovery.","is_dataset_classified":null,"base_score":5.337538079701318,"endowment":5.337538079701318,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"17629962","pmcid":"PMC2958218","openalex_id":"https://openalex.org/W2014862375","authors":[],"funders":[{"funder_name":"NIMH NIH HHS","grant_id":"MH073910","title":null},{"funder_name":"NIMH NIH HHS","grant_id":"MH040537","title":null},{"funder_name":"NIMH NIH HHS","grant_id":"R01 MH040537","title":null},{"funder_name":"NIMH NIH HHS","grant_id":"U19 MH082441","title":null},{"funder_name":"NIMH NIH HHS","grant_id":"R21 MH073910","title":null}],"total_grants":5,"fwci":8.4305,"citation_percentile":0.98560986,"influential_citations":0,"citation_trend":[{"year":2012,"count":28},{"year":2013,"count":13},{"year":2014,"count":13},{"year":2015,"count":11},{"year":2016,"count":9},{"year":2017,"count":7},{"year":2018,"count":9},{"year":2019,"count":7},{"year":2020,"count":3},{"year":2021,"count":5},{"year":2022,"count":3},{"year":2023,"count":1},{"year":2024,"count":2},{"year":2025,"count":4}],"oa_status":"green","license":"https://www.elsevier.com/tdm/userlicense/1.0/","oa_locations":[{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/2958218","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/2958218","host_type":"repository"},{"url":"https://api.elsevier.com/content/article/PII:S0165614707001423?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S0165614707001423?httpAccept=text/plain","host_type":"publisher"},{"url":"https://doi.org/10.1016/j.tips.2007.06.002","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/17629962","host_type":"repository"}],"fields_of_study":["Receptor Mechanisms and Signaling","Neuropeptides and Animal Physiology","Neurotransmitter Receptor Influence on Behavior","Animals","Drug Design","Humans","Models, Biological","Pharmaceutical Preparations","Receptors, G-Protein-Coupled","Signal Transduction","Technology, Pharmaceutical"],"mesh_terms":["Animals","Pharmaceutical Preparations","Humans","Models, Biological","Technology, Pharmaceutical","Drug Design","Signal Transduction","Receptors, G-Protein-Coupled"],"keywords":["Aripiprazole","Partial agonist","Functional selectivity","Neuroscience","In vivo","Agonist","Antipsychotic","Drug discovery","Pharmacology","Dopamine receptor D2","Intrinsic activity","Receptor","Biology","Psychology","Computational biology","Dopamine","Schizophrenia (object-oriented programming)","Bioinformatics","Biochemistry","Genetics","Psychiatry"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Good health and well-being"}],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-17T01:43:26.025967Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
