Allogeneic CD8 + T cells can be made from pluripotent stem cells (PSCs), but deriving CD4 + T cells from PSCs has remained a significant challenge. Using feeder- and serum-free conditions, we found that CD4 + vs. CD8 + T cell commitment from PSCs can be controlled by fine-tuning the dynamics of Notch and T cell receptor (TCR) signaling delivered to CD4 + CD8 + double-positive T cells. Notch signaling negatively impacts CD4 + T cell commitment, and its timed removal allows generation of clonally diverse and expandable CD4 + T cells from PSCs. The resulting CD4 + T cells respond to cytokine-mediated polarization by differentiating into Th1, Th2, or Th17 cells, recapitulating canonical helper cell function. These findings represent a significant step toward using PSC-derived CD4 + T cells as a low-cost, off-the-shelf cell therapy.","is_dataset_classified":null,"base_score":1.791759469228055,"endowment":1.791759469228055,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"41512836","pmcid":null,"openalex_id":"https://openalex.org/W7119483884","authors":[],"funders":[{"funder_name":"Michael Smith Health Research BC","grant_id":"RT-2021-1946","title":null},{"funder_name":"Canadian Institutes of Health Research","grant_id":"ICC-17644","title":null},{"funder_name":"Canadian Institutes of Health Research","grant_id":"FDN-159908","title":null},{"funder_name":"Canada Foundation for Innovation","grant_id":"38395","title":null},{"funder_name":"Genome British Columbia","grant_id":"SIP031","title":null},{"funder_name":"Natural Sciences and Engineering Research Council of Canada","grant_id":"unidentified","title":"unidentified"},{"funder_name":"British Columbia Knowledge Development Fund","grant_id":"","title":null},{"funder_name":"Wellcome Trust","grant_id":"","title":null},{"funder_name":"Natural Sciences and Engineering Research Council of Canada","grant_id":"","title":null},{"funder_name":"BC Children's Hospital","grant_id":"","title":null}],"total_grants":10,"fwci":24.4311,"citation_percentile":0.99481297,"influential_citations":0,"citation_trend":[{"year":2026,"count":5}],"oa_status":"hybrid","license":"cc-by-nc-nd","oa_locations":[{"url":"https://doi.org/10.1016/j.stem.2025.12.010","host_type":"journal"},{"url":"https://doi.org/10.1016/j.stem.2025.12.010","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S1934590925004448?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S1934590925004448?httpAccept=text/plain","host_type":"publisher"},{"url":"https://pubmed.ncbi.nlm.nih.gov/41512836","host_type":"repository"}],"fields_of_study":["CAR-T cell therapy research","Pluripotent Stem Cells Research","CRISPR and Genetic Engineering","0303 health sciences","03 medical and health sciences","Humans","Cell Differentiation","CD4-Positive T-Lymphocytes","CD8-Positive T-Lymphocytes","Pluripotent Stem Cells","Receptors, Notch","Signal Transduction","Receptors, Antigen, T-Cell"],"mesh_terms":["Cell Differentiation","Humans","Receptors, Antigen, T-Cell","Signal Transduction","CD4-Positive T-Lymphocytes","CD8-Positive T-Lymphocytes","Pluripotent Stem Cells","Receptors, Notch"],"keywords":["Induced pluripotent stem cell","T cell","Stem cell","Notch signaling pathway","Cell","Interleukin 21","Cytotoxic T cell","T cells","Pluripotent stem cells","T cell polarization","Notch signaling","Regenerative Medicine","Tcr Signaling","Helper T Cells","T Cell Development","Tcr Repertoires","Ipsc-t Cells","CD4-Positive T-Lymphocytes","Receptors, Notch","Receptors, Antigen, T-Cell","Humans","Cell Differentiation","CD8-Positive T-Lymphocytes","Signal Transduction"],"sdg_mappings":[],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-12T06:20:48.247131Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
