CDK activates origin firing by phosphorylating two substrates, Sld2 and Sld3, forming a transient and limiting intermediate—the pre-initiation complex (pre-IC). Here, we show in the budding yeast Saccharomyces cerevisiae that the CDK phosphorylations of Sld3 and Sld2 are rapidly turned over during S phase by the PP2A and PP4 phosphatases. PP2A Rts1 targets Sld3 specifically through an Rts1-interaction motif, and this targeted dephosphorylation is important for origin firing genome-wide, for formation of the pre-IC at origins and for ensuring that Sld3 is dephosphorylated in G1 phase. PP2A Rts1 promotes replication in vitro , and we show that targeted Sld3 dephosphorylation is critical for viability. Together, these studies demonstrate that phosphatases enforce the correct ordering of replication factor phosphorylation and in addition to kinases are also key drivers of replication initiation.","is_dataset_classified":null,"base_score":2.3978952727983707,"endowment":2.3978952727983707,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"36543171","pmcid":null,"openalex_id":"https://openalex.org/W4312083065","authors":[],"funders":[{"funder_name":"Wellcome Trust","grant_id":"107056/Z/15/Z","title":null},{"funder_name":"Wellcome Trust","grant_id":"092096","title":null},{"funder_name":"Cancer Research UK","grant_id":"C6946/A14492","title":null},{"funder_name":"Biotechnology and Biological Sciences Research Council","grant_id":"BB/M011194/1","title":null},{"funder_name":"Cancer Research UK","grant_id":" C15873/A12700","title":null}],"total_grants":5,"fwci":0.7843,"citation_percentile":0.68953837,"influential_citations":0,"citation_trend":[{"year":2023,"count":4},{"year":2024,"count":3},{"year":2025,"count":3}],"oa_status":"hybrid","license":"cc-by","oa_locations":[{"url":"http://www.cell.com/article/S1097276522011364/pdf","host_type":"journal"},{"url":"http://www.cell.com/article/S1097276522011364/pdf","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S1097276522011364?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S1097276522011364?httpAccept=text/plain","host_type":"publisher"},{"url":"https://doi.org/10.1016/j.molcel.2022.12.001","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/36543171","host_type":"repository"},{"url":"https://www.cell.com/article/S1097276522011364/pdf","host_type":"Unpaywall"}],"fields_of_study":["Mitochondrial Function and Pathology","Microtubule and mitosis dynamics","DNA Repair Mechanisms"],"mesh_terms":["DNA Replication","DNA-Binding Proteins","Saccharomycetales","Phosphorylation","Saccharomyces cerevisiae","Replication Origin","Cell Cycle Proteins","Cyclin-Dependent Kinases","Saccharomyces cerevisiae Proteins"],"keywords":["Biology","Dephosphorylation","Evolutionary biology","Phosphorylation","Computational biology","Cell biology","Phosphatase","Phosphatases","Cell cycle","DNA replication","Yeast","Genome Stability","Origin Firing"],"sdg_mappings":[],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-06T22:47:10.838745Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
