By permeabilizing Marseillevirus virions and performing genome-wide nuclease digestion, chemical cleavage, and mass spectrometry assays, we find that the higher-order organization of Marseillevirus chromatin fundamentally differs from that of eukaryotes. Marseillevirus nucleosomes fully protect DNA within virions as closely abutted 121-bp DNA-wrapped cores without linker DNA or phasing along genes. Likewise, we observed that nucleosomes reconstituted onto multi-copy tandem repeats of a nucleosome-positioning sequence are tightly packed. Dense promiscuous packing of fully wrapped nucleosomes rather than \"beads on a string\" with genic punctuation represents a distinct mode of DNA packaging by histones. We suggest that doublet histones have evolved for viral genome protection and may resemble an early stage of histone differentiation leading to the eukaryotic octameric nucleosome.","is_dataset_classified":null,"base_score":3.6375861597263857,"endowment":3.6375861597263857,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"36370708","pmcid":null,"openalex_id":"https://openalex.org/W4308787134","authors":[],"funders":[{"funder_name":"Howard Hughes Medical Institute","grant_id":"","title":null},{"funder_name":"David and Lucile Packard Foundation","grant_id":"","title":null},{"funder_name":"Howard Hughes Medical Institute","grant_id":"","title":null}],"total_grants":3,"fwci":6.2242,"citation_percentile":0.96373658,"influential_citations":0,"citation_trend":[{"year":2022,"count":3},{"year":2023,"count":6},{"year":2024,"count":13},{"year":2025,"count":10},{"year":2026,"count":5}],"oa_status":"hybrid","license":"cc-by","oa_locations":[{"url":"https://doi.org/10.1016/j.molcel.2022.10.020","host_type":"journal"},{"url":"https://doi.org/10.1016/j.molcel.2022.10.020","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S1097276522010206?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S1097276522010206?httpAccept=text/plain","host_type":"publisher"},{"url":"https://pubmed.ncbi.nlm.nih.gov/36370708","host_type":"repository"},{"url":"https://amu.hal.science/hal-04003911","host_type":"repository"}],"fields_of_study":["Plant Virus Research Studies","Bacteriophages and microbial interactions","Animal Virus Infections Studies","Nucleosomes","Histones","Giant Viruses","DNA","Virion","Genome, Viral"],"mesh_terms":["Giant Viruses","DNA","Histones","Nucleosomes","Virion","Genome, Viral"],"keywords":["Nucleosome","Biology","Histone","Linker DNA","DNA","Genome","Chromatin","Micrococcal nuclease","Genetics","Histone methylation","Cell biology","Gene","DNA methylation","Marseillevirus","Mnase-seq","Mpe-seq","Doublet Histones"],"sdg_mappings":[],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-10T10:13:58.961886Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
