The T-cell populations and mechanisms involved in clearance of virus from the genital epithelium were examined in this study. T lymphocytes expressing either alphabeta or gammadelta T-cell receptors (TCR) have been detected in the vaginal epithelium of mice. The involvement of gammadelta T cells in HSV-2 clearance was tested by intravaginal (ivag) challenge of mice depleted of alphabeta T cells by administration of specific antibodies and of mice lacking gammadelta T cells due to specific deletion of the delta TCR gene. The results of these studies strongly suggest that gammadelta T cells are not required for or involved in clearance of HSV-2 from the genital epithelium. Mechanisms of virus clearance employed by alphabeta T cells were also examined. Although HSV-specific lytic activity could be demonstrated ex vivo in populations of vaginal exudate cells from HSV-infected mice, clearance of virus did not require either perforin- or Fas/Fas ligand (FasL)-dependent cytolytic pathways. In contrast, virus resolution was significantly impaired following neutralization of interferon-gamma (IFN-gamma), but not tumor necrosis factor-alpha (TNF-alpha). Together, these results suggest that non-lytic mechanisms mediated by alphabeta T cells were responsible for resolution of a genital HSV-2 infection.","is_dataset_classified":null,"base_score":3.6888794541139363,"endowment":3.6888794541139363,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"15063634","pmcid":null,"openalex_id":"https://openalex.org/W1999098899","authors":[],"funders":[{"funder_name":"NIAID NIH HHS","grant_id":"AI-054444","title":null},{"funder_name":"NIAID NIH HHS","grant_id":"AI-42815","title":null}],"total_grants":2,"fwci":1.5293,"citation_percentile":0.82304219,"influential_citations":0,"citation_trend":[{"year":2012,"count":1},{"year":2014,"count":2},{"year":2015,"count":4},{"year":2016,"count":1},{"year":2018,"count":1},{"year":2021,"count":2},{"year":2023,"count":2},{"year":2024,"count":1},{"year":2025,"count":2}],"oa_status":"closed","license":"https://www.elsevier.com/tdm/userlicense/1.0/","oa_locations":[{"url":"https://api.elsevier.com/content/article/PII:S0165037803001578?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S0165037803001578?httpAccept=text/plain","host_type":"publisher"},{"url":"https://doi.org/10.1016/j.jri.2003.12.002","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/15063634","host_type":"repository"}],"fields_of_study":["Herpesvirus Infections and Treatments","Immune Cell Function and Interaction","Cytomegalovirus and herpesvirus research"],"mesh_terms":["Animals","Cells, Cultured","Epithelial Cells","Female","Herpes Genitalis","Interferon-gamma","Lymphocyte Activation","Membrane Glycoproteins","T-Lymphocytes, Cytotoxic","Tumor Necrosis Factor-alpha","Vagina","Receptors, Antigen, T-Cell, gamma-delta","Receptors, Antigen, T-Cell, alpha-beta","Cell Death","Herpesvirus 2, Human","Immunity, Mucosal","fas Receptor","Mice","Pore Forming Cytotoxic Proteins","Fas Ligand Protein","Perforin"],"keywords":["Herpes simplex virus","Genital herpes","HSL and HSV","Virology","Herpes Genitalis","Biology","Sex organ","Virus","Genetics"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Good health and well-being"}],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-02T23:04:13.655684Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
