{"doi":"10.1016/j.jmb.2025.169082","title":"The Evolution of ABC Importers","abstract":"• Structural and mechanistic differences between Type I and II Importers highlight distinct states and the complexity of transport cycles. • Variability in substrate binding proteins and presence of accessory domains adds an additional layer of functional plasticity to ABC importers. • Evidence of ABC importer-exporter overlap and domain swapping indicate transporter adaptability in cellular nutrient acquisition. • Understanding importer organization and dynamics within cells is essential for biological and biotechnological technological applications. I am an Associate Professor in the Department of Molecular Biosciences at Northwestern University. My research program investigates the structure, function, and regulation of membrane proteins, with a particular emphasis on ATP-binding cassette (ABC) importers. ABC transporters are a highly conserved superfamily of transmembrane proteins found across all organisms. These proteins utilize the energy from ATP binding and hydrolysis to transport of a broad array of substrates– including metabolites, lipids, peptides and drugs– across cellular membranes. In this perspective, I discuss how structural and biophysical characterization of ABC importers have significantly advanced our understanding of the mechanisms underlying their transport function. I also highlight the challenges in developing a unified mechanistic model and propose that the remarkable diversity of ABC transporters may necessitate multiple transport mechanisms for a complete picture of how these critical proteins function.","journal":"Journal of Molecular Biology","year":2025,"id":540616,"datarank":0.0,"base_score":0.0,"endowment":0.0,"self_citation_contribution":0.0,"citation_network_contribution":0.0,"self_endowment_contribution":0.0,"citer_contribution":0.0,"corpus_percentile":null,"corpus_rank":null,"citation_count":2,"citer_count":0,"citers_with_citation_signal":0,"citers_with_endowment":0,"datacite_reuse_total":0,"is_dataset":false,"is_dataset_confidence":0.9479,"is_data_producer":false,"deposit_databanks":null,"is_oa":true,"file_count":0,"downloads":0,"has_version_chain":false,"published_date":"2025-01-01","fair_score":null,"fair_percentile":null,"algorithm_id":"datarank_citation_only_1hop_v6","ranking_scope":"data_only","authors":[{"id":302437,"name":"Heather W.
