In patients affected by bipolar disorder rs4680 can influence antidepressant response and the propensity to develop psychotic symptoms, with the Met/Met genotype exerting a protective role. The same genotype could influence other dopamine-associated psychopathological features, such as mania.
Methods
We genotyped rs4680 in a sample of 163 patients affected by bipolar disorder type I, and assessed the personal history of recurrence of the illness.
Results
We observed a significant association between homozygosis for the rs4680 COMT low-activity variant and a reduced recurrence of manic, but not depressive, episodes during the course of the illness.
Conclusions
We suggest that rs4680 could be an inheritable aspect of the mechanisms of dopamine regulation that influence the individual susceptibility of patients with bipolar disorder to develop manic episodes of illness.","is_dataset_classified":null,"base_score":3.6109179126442243,"endowment":3.6109179126442243,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"21397335","pmcid":null,"openalex_id":"https://openalex.org/W2036575957","authors":[],"funders":[],"total_grants":0,"fwci":0.5049,"citation_percentile":0.64080164,"influential_citations":0,"citation_trend":[{"year":2012,"count":2},{"year":2014,"count":1},{"year":2015,"count":2},{"year":2016,"count":3},{"year":2017,"count":2},{"year":2019,"count":1},{"year":2021,"count":23},{"year":2022,"count":1},{"year":2023,"count":1}],"oa_status":"closed","license":"https://www.elsevier.com/tdm/userlicense/1.0/","oa_locations":[{"url":"https://api.elsevier.com/content/article/PII:S0165032711000760?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S0165032711000760?httpAccept=text/plain","host_type":"publisher"},{"url":"https://doi.org/10.1016/j.jad.2011.02.022","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/21397335","host_type":"repository"}],"fields_of_study":["Bipolar Disorder and Treatment","Neurotransmitter Receptor Influence on Behavior","Schizophrenia research and treatment"],"mesh_terms":["Adult","Alleles","Bipolar Disorder","Catechol O-Methyltransferase","Dopamine","Female","Genotype","Homozygote","Humans","Male","Middle Aged","Polymorphism, Genetic","Recurrence","Age of Onset","Genetic Predisposition to Disease"],"keywords":["rs4680","Catechol-O-methyl transferase","Mania","Bipolar disorder","Psychology","Psychiatry","Genotype","Tranylcypromine","Internal medicine","Medicine","Genetics","Chemistry","Mood","Gene","Monoamine oxidase","Biology"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Good health and well-being"}],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[{"name":"refsnp"}],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-01T09:38:46.515352Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
