Leptospirosis is a worldwide zoonotic disease caused by a pathogenic species of Leptospira and highly endemic in tropical regions which is becoming a main concern in public health and health care settings. Studies conducted on outbreak areas revealed that rodents are closely associated with leptospirosis transmission. Despite of the high number of human cases reported in Malaysia in recent year, the information regarding the primary vector and circulating serovar of Leptospira is still limited. Thus, the present study was undertaken to identify and characterize pathogenic Leptospira species among small mammals captured in wet markets via amplification of tissue sample and culture. Methods & Materials: The kidneys of 89 captured small mammals from wet markets were harvested. A portion of the kidneys was cultured into EMJH medium and incubated for 3 months while another portion was subjected to flaB gene amplification by polymerase chain reaction (PCR). Positive growth of leptospires was also subjected to flaB gene amplification for the detection of pathogenic species of Leptospira. Positive samples were then further characterized using MLST Scheme 1. Results: Thirty one samples (34.8%) were detected positive for pathogenic Leptospira, with 3 samples detected by culture, 16 samples by PCR and 12 by culture and PCR. The sequencing results of flaβ gene representative samples showed 90.3% is L. interrogans and 9.7% is L. borgepetersenii. From the MLST analysis, three clone of Leptopsira was identified namely L. interrogans serovar Bataviae ST50, L. interrogans serovar Bataviae ST205 and L. borgeptersenii serovar Javanica ST143. Similar species was also observed via MLST and flaB amplification. Conclusion: High numbers of pathogenic Leptospira was identified among small mammals captured indicate that there is a potential risk for the visitor and worker to acquire leptospirosis. Species identification is sufficient via flaB amplification with better Leptospira detection from tissue sample via direct amplification compared to culture.
